Clonidine Pretreatment Inhibits Stress-Induced Gastric Ulcer in Rats

Abstract: Stressful circumstances can cause stomach ulcers, which can bleed, exposing patients to potentially life-threatening complications. In the present animal study, we showed that clonidine, a routinely available medication, may be useful in preventing stress-induced stomach ulcers.

“…The high ulcer indices in rats treated with these ulcerogens in this study confirm the efficacy of these methods and the veracity of these reports. Involvement of oxygen‐derived free radicals in the pathogenesis of gastrointestinal mucosal damage induced by NSAIDs [23], ethanol [24–26], feeding restriction stress [27], and H. pylori [28,29] is well established. It has also been suggested that free radicals generated by neutrophils may be an important factor delaying the healing of acetic acid‐induced chronic gastric ulcer injury [16].…”

“…Central sympathetic activation was examined with the use of clonidine, a commonly applied pharmacological model; after an initial peripheral α 1 -stimulation, the central action of clonidine prevails, resulting in inhibition of sympathetic preganglionic neurons and decreased sympathetic drive ( 21 ). As previously ( 22 ), clonidine was given intraperitoneally (0.5 mg/kg), 1 h prior to the experiments.…”

“…The α 2-noradrenergic receptors and the α 2 presynaptic are involved in the regulation of gastric acid secretion and are effective in protection against chemical agents such as NSAIDs and ethanol, where their effects may be mediated central and peripheral receptors [47]. …”
