Clinical value of ELISA assays for IgM and IgG rheumatoid factors.

Abstract: SUMMARY The clinical value of enzyme linked immunosorbent assays (ELISA) assays for IgM and IgG rheumatoid factors was assessed in a series of studies using rabbit IgG as antigen. The tests were reproducible with intra-assay coefficients of variation of 6% and could be simply and rapidly performed. Normal ranges were established using 106 sera from healthy controls. In a cross sectional study of 208 rheumatoid patients these assays were compared with the Rose-Waaler and laser nephelometric assessments of rheum… Show more

“…The estimated inter-assay %CV’s were 15% for FMIA ( n = 8) and 15% for ELISA ( n = 16). These values are consistent with inter-assay %CV’s reported for other IgM assays (11–13% [ 23 ], <20% [ 24 ], 8% [ 25 ], 14–21% [ 26 ]). Signals from dilution series (1:100, 1:200, 1:400, 1:800) for eight samples tested by FMIA and ELISA were normalized to fall on a single linear response curve for each method ( Supplementary Figure S4 ).…”

“…CCP ELISAs have intra-assay CVs ranging from 4.8% to 13% and inter-assay CVs ranging from 9% to 17%, 15 while RF ELISAs have an intra-assay CV of 6% and an inter-assay CV of 8%. 16 …”
