Clinical utility of cycle threshold values in the context of COVID-19

Abstract: BackgroundThe ability to predict likely prognosis and infectiousness for patients with COVID-19 would aid patient management decisions. Diagnosis is usually via real-time PCR and it is unclear whether the semi-quantitative capability of this method, determining viral load through cycle threshold (Ct) values, can be leveraged.ObjectivesWe aim to review available knowledge on correlations between SARS-COV-2 Ct values and patient- or healthcare-related outcomes to determine whether Ct values provide useful clinic… Show more

“…The cycle threshold (Ct) values of RT-PCR represent the number of amplification cycles that are needed by the target gene to surpass a certain threshold level. Ct values are inversely proportional to viral load and can be used as an indirect way of quantifying viral RNA copy numbers [ 4 ].…”

“…The disease COVID-19, caused by infection with SARS-CoV-2, is associated with a series of physiopathological mechanisms that mobilize a wide variety of biomolecules, especially immunological ones (Costela-Ruiz et al, 2020;Li et al, 2020). The gold standard for infection diagnosis is real-time reverse transcriptase PCR (RT-PCR) (Kevadiya et al, 2021;Rao et al, 2020;Younes et al, 2020). Following the emergence of the COVID-19 pandemic, D-dimer was established as a possible predictor of prognosis for COVID-19 patients.…”

“…3 Cycle threshold values of real-time polymerase chain reaction (PCR), which is the gold-standard diagnostic method for COVID-19, are representative of assumed viral loads in the tested specimen. 4 The role of viral loads in COVID-19 severity, mortality, and transmission is still a matter of question.…”
