Clinical pharmacology of tramadol and tapentadol, and their therapeutic efficacy in different models of acute and chronic pain in dogs and cats

Abstract: Opioids are considered the gold standard to manage acute or chronic or mild to severe pain. Tramadol is a widely prescribed analgesic drug for dogs and cats; it has a synthetic partial agonism on μ-opioid receptors and inhibits the reuptake of norepinephrine and serotonin. However, the biotransformation and resultant metabolites differ between species and depend on cytochrome P450 interactions. Dogs mainly produce the inactive N-desmethyl tramadol metabolite, whereas cats exhibit an improved antinociceptive ef… Show more

“…They mention that less analgesia was provided in the groups where monotherapy drugs were used than in the combination of two analgesics (Garcia-Pereira 2018). Tramadol, used in rescue analgesic, is a drug with a serotonergic activity that provides similar analgesia to that reported when administering pure opioids during surgical procedures such as OVH in bitches (Mastrocinque and Fantoni 2003;Zhang et al 2017;Oliva-Domínguez et al 2021). In this sense, in some studies, they observed that in the animals requiring analgesic rescue, tramadol proved effective for controlling acute pain in dogs despite having received non-steroidal analgesics prior to surgery (Imagawa et al 2011;Teixeira et al 2013).…”

“…Due to the adverse effects that NSAIDs can generate, especially for treating chronic afflictions such as arthritis and cancer, opioids are another therapeutic option [ 101 ]. However, since the long-term use of these drugs is also associated with complications, research has begun to new concepts and explore directions.…”

“…Adverse events were dose dependent, and caution should be exercised in cats that have concurrent disease or are receiving other drugs that may produce adverse gastrointestinal effects. Side effects of gabapentin use include mydriasis, sedation, euphoria and vomiting, and the bitter taste can make this treatment unpalatable to both cats and owners (Domínguez‐Oliva and others 2021).…”
