Clinical Evaluation of Kinetic Enzymatic Fixed-Time and Integral Analysis of Serum Triglycerides

Abstract: Serum triglycerides can be enzymatically determined by hydrolysis of triglycerides to glycerol and free fatty acids [Clin. Chem. 19, 476 (1973)]. Enzyme integral (end-point) and fixed-time rate analyses of serum triglycerides by this procedure have been investigated with a centrifugal analyzer. We determined the precision of blank determinations, the contribution of normal serum alkaline phosphatase to total values, and the amount of endogenous glycerol in fasting serum samples and blood collection tubes. We a… Show more

“…TG are converted to free glycerol by lipase, and the glycerol is oxidized (by glycerol dehydrogenase) to dihydroxy-acetone under formation of NADH, which is measured using a kinetic, bichromatic method. Preexisting glycerol (usually below 0.1 mmol/L) is included in the result (28).…”

“…The diabetic rabbits exhibiting persistent blood sugar level in the range of 250-300 mg/dl were selected to determine efficacy of the formulation. Alloxan was used since it produce hyperglycemia ,hypercholesterolemia and elevated lipid peroxidases levels [19] which is the appropriate model for the present study.…”

“…The plasma was separated from the cells and stored in LP tubes at a temperature of -20 o C until analysis. The fasting plasma cholesterol and triglyceride levels were determined by enzymatic method (Roschkau et al, 1975;Sidel et al, 1981) and (Tiffany et al, 1974) respectively using Boehringer-Mannheim kit method.…”
