magnetic field. From the discovery of electromagnetic induction to the quantum Hall effect, humans have strived for seeking applicability of novel mechanisms in the magneto-sensors. [2] Currently, the magneto-sensors are generally based on the quantum effects, which relies on a highly sensitive interface of semiconductor materials commonly fabricated by epitaxial growth. [3] With development of flexible electronics and bioelectronics, these semiconductor based technics seem increasingly unsuitable for the biomedical applications.Interestingly, some species possess an amazing capability of magneto-perception such as migratory birds, [4] butterflies, [5] and honeybees. [6] These animals perceive the geomagnetic field relying on special proteins. Cryptochrome (Cry) has been considered for a long time to mediate the magneto-perception via quantum spin dynamics of light-induced radical-pair, which are excited by sunlight and can be modulated by the geomagnetic field. [7][8][9] This principle was employed to sense the magnetic field because the life-time and the spectral characteristic of radical pair in singlet and triplet states can reflect the information about magnetic field. [10] Cry played an important role in the During billions of years of evolution, some species develop fantastic magnetoperception, offering a biomimetic route to develop the next generation of magneto-sensors. Recently, a novel principle was proposed to explain the navigation of pigeons in the presence of geomagnetic fields and sunlight. The key link is thought to lie in the magneto-optically involved conformational variation of magnetoreceptor protein (MagR)/cryptochrome (Cry4) complex. The MagR/Cry4 complex is fabricated and purified in vitro, creating a magnetosensing device by immobilization of this protein on a graphene-modified electrochemical electrode. By using electrochemical impedance spectroscopy, magneto-sensing with a current detectability of 10 mT is realized with MagR/ Cry4 complex. It is proved that this process requires the involvement of both magnetic fields and light, partly confirming in vitro the magneto-perceptive mechanism of MagR/Cry4 complex. It is also shows that this device can be used to reflect the anisotropic responses of nanomaterials to the external magnetic field. It is believed this protein-based magneto-sensing will greatly boost development of bioelectronics and deepen understanding of the phenomena of magneto-perception for organisms.

The global emergence of coronavirus disease 2019 (COVID‐19) has caused substantial human casualties. Clinical manifestations of this disease vary from asymptomatic to lethal, and the symptomatic form can be associated with cytokine storm and hyperinflammation. In face of the urgent demand for effective drugs to treat COVID‐19, we have searched for candidate compounds using in silico approach followed by experimental validation. Here we identified celastrol, a pentacyclic triterpene isolated from Tripterygium wilfordii Hook F, as one of the best compounds out of 39 drug candidates. Celastrol reverted the gene expression signature from severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2)‐infected cells and irreversibly inhibited the recombinant forms of the viral and human cysteine proteases involved in virus invasion, such as M pro (main protease), PL pro (papain‐like protease), and recombinant human cathepsin L. Celastrol suppressed SARS‐CoV‐2 replication in human and monkey cell lines and decreased interleukin‐6 (IL‐6) secretion in the SARS‐CoV‐2‐infected human cell line. Celastrol acted in a concentration‐dependent manner, with undetectable signs of cytotoxicity, and inhibited in vitro replication of the parental and SARS‐CoV‐2 variant. Therefore, celastrol is a promising lead compound to develop new drug candidates to face COVID‐19 due to its ability to suppress SARS‐CoV‐2 replication and IL‐6 production in infected cells.

Comparative study of cigarette smoke, Klebsiella pneumoniae, and their combination on airway epithelial barrier function in mice

Background: The airway epithelium acts as a physical barrier to protect pulmonary airways against pathogenic microorganisms and toxic substances, such as cigarette smoke (CS), bacteria, and viruses. The disruption of the structural integrity and dysfunction of the airway epithelium is related to the occurrence and progression of chronic obstructive pulmonary disease.Purpose: The aim of this study is to compare the effects of CS, Klebsiella pneumoniae (KP), and their combination on airway epithelial barrier function. Methods:The mice were exposed to CS, KP, and their combination from 1 to 8 weeks. After the cessation of CS and KP at Week 8, we observed the recovery of epithelial barrier function in mice for an additional 16 weeks. To compare the epithelial barrier function among different groups over time, the mice were sacrificed at Weeks 4, 8, 16, and 24 and then the lungs were harvested to detect the pulmonary pathology, inflammatory cytokines, and tight junction proteins. To determine the underlying mechanisms, the BEAS-2B cells were treated with an epidermal growth factor receptor (EGFR) inhibitor (AG1478). Results:The results of this study suggested that the decreased lung function, increased bronchial wall thickness (BWT), elevated inflammatory factors, and reduced tight junction protein levels were observed at Week 8 in CS-induced mice and these changes persisted until Week 16. In the KP group, increased BWT and elevated inflammatory factors were observed only at Week 8, whereas in the CS + KP group, decreased lung function, lung tissue injury, inflammatory cell infiltration, and epithelial
