“Click” Immobilization of a VEGF-Mimetic Peptide on Decellularized Endothelial Extracellular Matrix to Enhance Angiogenesis

Abstract: We show that coating of decellularized extracellular matrix (DC-ECM) on substrate surfaces is an efficient way to generate a platform mimicking the native ECM environment. Moreover, the DC-ECM can be modified with a peptide (QK) mimicking vascular endothelial growth factor without apparently compromising its integrity. The modification was achieved through metabolic incorporation of a “clickable” handle to DC-ECM followed by rapid attachment of the QK peptide with an azido tag using copper-catalyzed click reac… Show more

“…For this reason, QK has high affinity to bind to VEGF receptors on the surface of endothelial cells [38,116]. Many studies have revealed that peptide QK promotes endothelial cell migration, viability, and proliferation in vitro [129][130][131][132][133] as well as supports angiogenesis in vivo [130,133,134].…”

“…16 The role of other ECM- and GF-derived peptides for directing cell behavior for vascularization are still being defined, including the use of a peptide that mimics vascular endothelial growth factor to direct angiogenesis. 9, 58, 60, 119 Also, peptide-driven self-assembly/gelation has been proposed as an improved bioink for organotypic culture and shown to be effective for in vivo angiogenesis. 17, 61-64, 79 Based on current trends, peptide modified polymers can be envisioned as a future direction for programming cell-specific bioink in 3D constructs to direct cell behavior for vascular patterning.…”

“…The tube formation assay is a widely-applied tool to detect cell angiogenesis [6,47]. Matrigel (BD Biosciences, USA) was added in the 96-well plates and allowed to polymerize for 30 min at 37℃.…”
