Esophageal squamous cell carcinoma (ESCC) is a lethal malignancy. However, there are few useful markers for diagnosis and treatment. Glutathione S‐transferase Pi 1 (GSTP1) has been reported as a predictor of malignancy or anticancer drug resistance in some cancers. We investigated the association of GSTP1 expression with the malignancy or drug resistance in ESCC cell lines and clinical tissue samples. Proliferation and apoptosis assays regarding GSTP1 expression were examined in ESCC cell lines. Proliferation of GSTP1 knockdown cells was significantly decreased (P < .01), and the frequency of early apoptosis was increased (P < .05). Invasion capacity of GSTP1 knockdown cells was slightly decreased in transwell assay. These results suggest that GSTP1 plays an important role in malignant potential. To examine the effects of GSTP1 on drug resistance, chemosensitivity assay and apoptosis assay under cisplatin exposure were carried out. Viability of GSTP1 knockdown cells treated with cisplatin was lower than that of control cells (P < .01). Moreover, the frequency of early and late apoptosis in GSTP1 knockdown cells was markedly increased over that of control cells by cisplatin exposure (P < .01). In immunohistochemistry assay of resected tissue samples, GSTP1 expression was significantly associated with clinical downstaging (P = .04) in 72 ESCC patients with neoadjuvant chemotherapy. Furthermore, there was a significant association between GSTP1 expression in resected tissue and biopsy samples in 34 ESCC patients without neoadjuvant chemotherapy (P = .02). In summary, GSTP1 was related to malignant potential and may be a predictive marker of drug resistance in ESCC patients.

The analysis of publications from the last 5 years is presented in this review. These articles describe the relationship of glutathione transferase (GST) with mammary tumors. Most of these works are dedicated to investigating the genotypic relationship between GST and neoplasms in human. The most common are single nucleotide polymorphism of GSTP1 Val105Ile (rs1695) and gene deletions GSTM1 and GSTT1. Several publications describe polymorphisms of GSTM3(rs4970737), GSTM4 and GSTA5. These polymorphisms (especially GSTP1 Val105Ile) are associated with the risk of mammary tumors, overall survival and relapse in patients of some ethnic groups. This is as a result of reduced enzymatic activity of GST and disturb of ability to detoxify substrates. Some studies demonstrate that single nucleotide polymorphism of GSTP1 Val105IIe is associated with a better response to chemotherapy and overall survival of patients, but no association with genotypes of GSTM1 and GSTT1. The injections of carcinogenic compounds into the tissue of mammary gland or its feeding to rats leed to decrease of GST activity and to development of tumors. The increased expression of GST in breast cancer cell lines leeds to increase of the resistance of these cells against various chemical compounds therefore the enzyme catalyzes the binding of these compounds to glutathione and this prevent their negative effects and is necessary for the formation of conjugates. Such increased expression of GST indicates on the resistance of breast cancer cell lines in particular to the action of drugs, which reduces their therapeutic effect. This is shown on cell lines BT474, MCF-7, MCF-7/ADR, MCF-7/ADR-1024, MCF-10A, MDA-MB-231, MDA-MB-468 and T47D. Chemical compounds (including antitumor drugs), that reduce the activity and/or expression of GST, can have a cytotoxic effect on these cells. Mammary tumors associated with epigenetic changes that do not change the sequence of nucleotides in GST genes. The methylation of the GSTP1 promoter decreases an expression of protein in mammary tissue and increases a risk of cancer in different ethnic groups. Compounds increase the level of expression of genes when they are able to lower the level of methylation or to affect demethylation. Polymorphisms of GST genes, their association with response to chemotherapy and overall survival of patients, expression of GST and it dependence on the action of anticancer drugs, methylation of GST promoter are associated with diagnosis, prognosis, and treatment of mammary neoplasms.

Background Breast cancer is the leading cause of death among women. Cisplatin is an effective drug for breast cancer, but resistance often develops during long term chemotherapy. While the mechanism of chemotherapy resistance is still not fully understood. Methods Survival analyses of ATP7A and ATP7B were used to evaluate their effects on the development of Breast invasive carcinoma (BRCA). Immunostaining, flow cytometry, and IC50 assay were utilized to examine the effects of ATP7A‐siRNA combined with cisplatin on apoptosis in breast cancer cells. Q‐PCR, western blotting, and dual‐luciferase assay were employed to confirm ATP7A is a novel target gene of miR‐148a‐3p. Results In this current study, we identified knocking‐down ATP7A could enhance cytotoxicity treatment of cisplatin in breast cancer cells. We also demonstrated miR‐148a‐3p overexpression in BRCA cells increased the sensitivity to cisplatin, and subsequently enhanced DNA damage and apoptosis. Moreover, we found ATP7A is a novel target gene of miR‐148a‐3p. In brief, our results showed miR‐148a could accelerate chemotherapy induced‐apoptosis in breast cancer cells by inhibiting ATP7A expression. Conclusions Our results highlight that inhibition of ATP7A is a potential strategy for targeting breast cancer resistant to cisplatin, and we provided an interesting method to compare the involvement of various genes in the assessment of cisplatin resistance.
