Classification of pyroptosis patterns and construction of a novel prognostic model for prostate cancer based on bulk and single-cell RNA sequencing

Abstract: BackgroundEmerging evidence suggests an important role for pyroptosis in tumorigenesis and recurrence, but it remains to be elucidated in prostate cancer (PCa). Considering the low accuracy of common clinical predictors of PCa recurrence, we aimed to develop a novel pyroptosis-related signature to predict the prognosis of PCa patients based on integrative analyses of bulk and single-cell RNA sequencing (RNA-seq) profiling.MethodsThe RNA-seq data of PCa patients was downloaded from several online databases. PCa… Show more

“…They constructed a prognostic signature based on these PRGs, their expression profile, a clinical database that can be used to precisely predict BCR after radical intervention, which is a determining risk factor for PCa specificity and distant metastasis, and the progression-free survival (PFS) rate [87]. Other researchers who repeated this work verified that the identified PRGs were closely related to carcinogenesis, tumor cell invasion, and the immune microenvironment in PCa [88,89].…”

“…The authors established a novel prognostic gene signature for predicting the recurrence free survival in PCa patients, which was shown to be an independent factor associated with the recurrence free survival in PCa. [ 103 ] Xiao et al established a prognostic risk model using a 6-hub PRG. By analyzing its relationship with PCa in terms of clinicopathological characteristics, tumor immune microenvironment and response to drug treatment, it was found that this risk model was of good clinical significance.…”

“…Pyroptosis appears to play a significant role in tumor progression and is linked to proliferation, migration, cell cycle, and treatment resistance in various of cancers, according to accumulated evidence ( Heo et al, 2019 ; Yu et al, 2019 ; Tan et al, 2021 ). Recent studies have found that pyroptosis-related genes (PRGs) have satisfactory predictive abilities in the prognosis of PCa and could be used as novel tumor biomarkers ( Fu et al, 2022 ; Hu et al, 2022 ; Wang et al, 2022 ). Meanwhile, its relationship with PCa immunity may provide assistance in the treatment of PCa ( Li et al, 2022 ; Zhang et al, 2022 ).…”
