Classification of CEA‐related positivity in primary and metastatic malignant melanoma

Abstract: Using a panel of antibodies to carcinoembryonic antigen (CEA), in paraffin-processed biopsy material patchy, predominantly membranous positivity was seen on tumour cells in 70 per cent of cases of superficial spreading melanoma, 60 per cent of nodular melanomas, and 75 per cent of secondary deposits studied with unabsorbed polyclonal anti-CEA only. No staining was seen using monoclonal anti-CEAs. Localization of CEA to the cell membrane was confirmed with confocal microscopy. Immunoblotting of fresh frozen mat… Show more

“…58 However, CEA can be shed by phospholipases from the cell surface through cleavage of its glycosylphosphatidylinositol-linkage, which causes the protein to be released in the circulation. This soluble CEA (sCEA) might therefore interfere with antibody based therapies.…”

“…1 Expression of diVerent families of adhesion molecules has been reported previously in melanoma, [2][3][4] with major changes reported in the progression of normal melanocytes to benign naevi. Further dramatic changes also occur when cells progress from dysplastic naevi or radial growth phase melanoma to biologically late vertical growth phase primary melanoma.…”

“…Further dramatic changes also occur when cells progress from dysplastic naevi or radial growth phase melanoma to biologically late vertical growth phase primary melanoma. [2][3][4][5][6] There is now compelling evidence to suggest that cadherins play a major role in epithelial cell-cell adhesion, with additional key roles recognised for cell phenotype, diVerentiation, epithelial/mesenchymal transformation, and invasion. [7][8][9] Loss of E-cadherin is associated with high grade morphology of tumours and there is a significant inverse relation between E-cadherin expression and survival in some tumours.…”

“…However, in independent studies conducted by Sanders et al [33] and Ravindranath et al [34] they found that anti cEA specific mAbs were not reactive with malignant melanoma. By contrast, Selby et al [35] found cEA immunoreactivity with a polyclonal antibody in a significant number of malignant melanoma cases.…”
