CITRX thioredoxin interacts with the tomato Cf-9 resistance protein and negatively regulates defence

Abstract: To identify proteins involved in tomato Cf-9 resistance protein function, a yeast two-hybrid screen was undertaken using the cytoplasmic C-terminus of Cf-9 as bait. A thioredoxin-homologous clone, interacting specifically with Cf-9, was identified and called CITRX (Cf-9-interacting thioredoxin). Virus-induced gene silencing (VIGS) of CITRX resulted in an accelerated Cf-9/Avr9-triggered hypersensitive response in both tomato and Nicotiana benthamiana, accompanied by enhanced accumulation of reactive oxygen spec… Show more

“…Ascorbate-deficient mutants exhibit microlesions and constitutive PR gene expression, lack induction of H 2 O 2 -sensitive genes, and display higher tolerance to infection by P. syringae when compared to wild type (Pavet et al, 2005). Silencing of the tobacco thioredoxin CITRX resulted in enhanced pathogen-induced ROS accumulation, calcium-dependent protein kinase activation, and defense gene expression (Rivas et al, 2004). Moreover, there is a possible implication of the cytosolic thioredoxin TRXh5 in response to pathogens and to oxidative stresses (Laloi et al, 2004).…”

“…In one report, virus-induced gene silencing of the cytosolic TRX-like protein CITRX convincingly demonstrated its implication in the Cf9/Avr9-triggered hypersensitive response (Rivas et al, 2004). The alternative NADPH-dependent redox pathway, which implicates glutaredoxins, is a potential source of redundancy.…”
