SummaryPlants employ cell surface receptors to recognize pathogen (or microbe)-associated molecular patterns (PAMPs/MAMPs), which are crucial for immune system activation. The well-studied Arabidopsis thaliana ELONGATION FACTOR-TU RECEPTOR (EFR) recognizes the conserved bacterial PAMP EF-Tu, and the derived peptides elf18 and elf26. The interfamily transfer of EFR has been shown to increase disease resistance in several crops, such as tomato, rice, wheat, and potato. Here, we generated sweet orange (Citrus sinensis) transgenic lines expressing EFR to test if it would confer broad-spectrum resistance against two important citrus bacterial diseases: citrus canker and citrus variegated chlorosis (CVC). Independent EFR transgenic lines gained responsiveness to elf18 and elf26 peptides from Xanthomonas citri and Xylella fastidiosa, as measured by reactive oxygen species (ROS) production, mitogen-activated protein kinase (MAPK) activation and defense gene expression. Consistently, infection assays showed that Citrus-EFR transgenic plants were more resistant to citrus canker and CVC. Our results show that the EFR immune receptor can improve plant immunity in a perennial crop against bacterial pathogens, opening perspectives to engineer durable broad-spectrum disease resistance under field conditions.

Citrus is one of the most important fruit crops in the world. This review will discuss the recent findings related to citrus transformation and regeneration protocols of juvenile and adult explants. Despite the many advances that have been made in the last years (including the use of inducible promoters and site-specific recombination systems), transformation efficiency, and regeneration potential still represent a bottleneck in the application of the new breeding techniques in commercial citrus varieties. The influence of genotype, explant type, and other factors affecting the regeneration and transformation of the most used citrus varieties will be described, as well as some examples of how these processes can be applied to improve fruit quality and resistance to various pathogens and pests, including the potential of using genome editing in citrus. The availability of efficient regeneration and transformation protocols, together with the availability of the source of resistance, is made even more important in light of the fast diffusion of emerging diseases, such as Huanglongbing (HLB), which is seriously challenging citriculture worldwide.

Valorization of Orange Peel Waste Using Precomposting and Vermicomposting Processes

The industrialization process of oranges generates waste, which is inadequately disposed of; this produces adverse effects on the environment. Among the alternatives for valorization is the vermicomposting process, which consists of the degradation of organic waste through the action of earthworms and microorganisms. Therefore, this research aimed to study this process using orange peel (OP) waste at the laboratory level. For this purpose, it was necessary to determine the degradation conditions through the monitoring of physicochemical parameters (temperature, pH, humidity, organic matter (OM), total organic carbon (TOC), total nitrogen (TN) and the carbon/nitrogen (C/N) ratio). To balance the substrate’s nutrients, load material (LM) that included vegetable waste and eggshells was added to three different mixtures: M1 (50% OP + 50% LM), M2 (40% OP + 60% LM) and M3 (60% OP + 40% LM). To condition the substrate for earthworm (Eisenia fetida) activity, a previous precomposting process was performed. The results showed that all the mixtures fulfilled the requirements for a quality and mature vermicompost; however, the highest concentrations for TN were in the mixtures M1 and M2. The total time required for degradation of the OP waste was 13 weeks.
