Abstract. Hepatocellular carcinoma (HCC) is one of the most common types of cancer across the world. Hyaluronic acid (HA) has been reported to serve an important role in tumor extension, progression, migration and invasion. In addition, the receptor for HA-mediated motility (RHAMM) has been demonstrated to be overexpressed in different types of cancer. However, whether the upregulation of RHAMM contributes to hepatocarcinogenesis of HCC remains unclear. The present study examined the RHAMM expression in 187 HCC patients by reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and immunohistochemistry (IHC). RHAMM expression was significantly upregulated in liver cancer tissues compared with that observed in adjacent normal liver tissues. The IHC analysis demonstrated that RHAMM was overexpressed in 18 (72.0%) of the 25 HCC tissues. Furthermore, overexpression of RHAMM was associated with tumor-node-metastasis (TNM), the presence of vascular invasion and recurrence. Notably, the present study indicated that the overexpression of RHAMM was closely associated with the shorter disease-free and overall survival, so it may be a potential independent predictor for disease-free and overall survival of HCC patients. In conclusion, the upregulation of RHAMM is associated with HCC progression and prognosis; and it may be a potential independent predictor of disease-free and overall survival of HCC following surgical resection. IntroductionLiver cancer is one of the most common neoplasms worldwide, and hepatocellular carcinoma (HCC) is the most prevalent type of liver cancer and accounts for 70-85% of all cases (1), ranking the sixth most common malignant tumors worldwide (2). More than 600,000 people lose their lives due to HCC every year (3), and HCC is the third most frequent cause of cancer-associated mortality (4). Previous studies have demonstrated that the incidence rate of HCC has been increasing. Surgical resection and liver-transplantation are superior therapeutic methods for HCC compared with other treatments, including radiofrequency ablation (RFA), transarterial chemoembolization (TACE) and chemotherapy, the treatment outcomes remain unsatisfactory owing to the high recurrence rate and high fatality rate of HCC (2). If HCC was detected and diagnosed earlier, surgical resection and liver-transplantation would result in an improved curative effect, therefore improving the disease-free and overall survival. The serum levels of α-fetoprotein (AFP) and ultrasonography (US) are widely used to screen for and assess HCC throughout the world. However, the sensitivity and specificity of AFP for HCC diagnosis and surveillance have certain limitations; US depends on the operator's skill, and clinicians are not always able to distinguish HCC clearly from other nodules (5). Circulating microparticles (MPs) are novel potential biomarkers in cancer patients. Their role in hepatocellular carcinoma (HCC) is under intensive investigation. In this study we tested the hypothesis that MPs expressing the antigen HepPar1 are increased in the blood of subjects with HCC and may serve as markers of early recurrence after liver resection (LR). We studied fifteen patients affected by HCC undergoing LR and used flow cytometry to assess the number of circulating HepPar1+ MPs. Ten subjects without HCC (five with liver cirrhosis and 5 with healthy liver) were used as controls. After LR, HCC patients were followed-up for early recurrence, which occurred in seven cases. The number of circulating HepPar1+ MPs was significantly higher in subjects affected by HCC, compared to individuals without cancer (p<0.01). We also found that, among HCC patients, the number of circulating HepPar1+ MPs, measured before LR, was significantly higher in those who displayed early recurrence compared to those without recurrence (p=0.02). Of note, other types of circulating MPs, such as those derived from endothelial cells (CD144+) or those produced by the activated endothelium (CD144+/CD62+) were not associated with HCC, neither could predict HCC recurrence. HepPar1+ MPs deserve further investigation as novel biomarkers of disease and prognosis in HCC patients.

Background Melanoma skin cancer remains the leading cause of skin cancer related deaths. Spitz lesions represent a subset of melanocytic skin lesions characterized by epitheloid or spindled melanocytes organized in nests. These lesions occupy a spectrum ranging from benign Spitz and atypical Spitz lesions all the way to malignant Spitz tumors. Appropriate management is reliant on accurate diagnostic classification, yet this effort remains challenging utilizing current light microscopic techniques. The discovery of novel biomarkers such as microRNAs (miR) may ultimately be a useful diagnostic adjunct for the evaluation of Spitz lesions. miR expression profiles have been suggested for non-Spitz melanomas but have yet to be ascribed to Spitz lesions. We hypothesized that distinct miR expression profiles would be associated with different lesions along the Spitz spectrum. Materials and Methods RNA extracted from paraffin embedded, formalin fixed tissues of 11 resected skin lesions including benign nevi (n=2), benign Spitz lesions (n=3), atypical Spitz lesions (n=3), and malignant Spitz tumors (n=3) were analyzed by the NanoString platform for simultaneous evaluation of over 800 miRs in each patient sample. Results Benign Spitz lesions had increased expression of miR-21-5p and miR-363-3p compared to benign nevi. Malignant Spitz lesions exhibited over-expression of miR-21-5p, miR-155-5p, and miR-1283 relative to both benign nevi and benign Spitz tumors. Notably, atypical Spitz tumors had increased expression of miR-451a and decreased expression of miR-155-5p expression relative to malignant Spitz lesions. Conversely, atypical Spitz lesions had increased expression of miR-21-5p, miR-34a-5p, miR-451a, miR-1283, and miR-1260a relative to benign Spitz tumors. Conclusions Overall, distinct miR profiles are suggested among Spitz lesions of varying malignant potential with some similarities to non-Spitz melanoma tumors. This work demonstrates the feasibility of this analytic method and forms the basis for further validation studies.
