Circulating humanin levels are associated with preserved coronary endothelial function

Abstract: Humanin is a small endogenous antiapoptotic peptide, originally identified as protective against Alzheimer's disease, but subsequently also found on human endothelium as well as carotid artery plaques. Endothelial dysfunction is a precursor to the development of atherosclerotic plaques, which are characterized by a highly proinflammatory, reactive oxygen species, and apoptotic milieu. Previous animal studies demonstrated that humanin administration may improve endothelial function. Thus the aim of this study w… Show more

“…Similar percentages of the 47 ED subjects and 33 control subjects were smokers, had hypertension and/or diabetes, and were on statin and/or aspirin therapy. As previously reported, levels of inflammatory markers (hs-CRP, P= 0.95) and traditional lipid CVD risk factors—including HDL-C ( P= 0.052), LDL-C ( P= 0.64), triglycerides ( P= 0.14), and Lp(a) ( P= 0.62)—did not differ significantly between the control and ED groups 3538 .…”

“…HN is expressed in the endothelial cell layer of human arteries and veins [7] and in carotid atherosclerotic plaques [8]. HN has also been detected in human skeletal muscle [9], plasma [10], testis and sperm [11]. HNr was shown to be expressed in a number of adult rat tissues such as cerebral cortex, hippocampus, heart, testis and uterus, among others [12].…”

“…The study protocol has been described in detail elsewhere (12,16,20,21). In brief, patients presenting to the catheterization laboratory with nonobstructive CAD (<40% stenosis) underwent invasive coronary microvascular functional testing using an intracoronary Doppler guidewire.…”

“…Impaired endothelial-independent microvascular function was defined as a coronary flow velocity reserve ratio in response to adenosine of #2.5 (22). Impaired endothelial-dependent microvascular function was defined as a maximal percentage increase in CBF in response to any dose of acetylcholine compared with baseline of #50% (16,17,21). A decrease in coronary artery diameter of $20% in response to acetylcholine compared with baseline was in keeping with epicardial endothelial dysfunction (16,17,21 Figure 1 There was a wide distribution of measurements of coronary flow velocity reserve (CFR) in response to adenosine and coronary blood flow (CBF) in response to acetylcholine among all patients presenting with chest pain and nonobstructive coronary artery disease.…”
