Circular RNAs in nucleus pulposus cell function and intervertebral disc degeneration

Abstract: Intervertebral disc degeneration (IDD) is a common cause of low back pain, which inflicts more global disability than any other condition. Although IDD was deemed to be a natural process that comes with ageing, a growing body of evidence suggested that both genetic and environmental factors could modify the development of IDD. In this connection, aberrant function of nucleus pulposus cells has been implicated in IDD pathogenesis. Circular RNAs are a novel class of endogenous non‐coding RNAs that play crucial r… Show more

“…A large of studies shown that the degeneration mainly displays as a large number of the nucleus pulposus cell apoptosis and thus nucleus pulposus cells apoptosis plays a crucial role in IVDD [7,8]. TNF-α is a key proinflammatory cytokine and could induce the apoptosis of nucleus pulposus cells [9].…”

“…CircVMA21 obviously inhibited inflammatory cytokines-induced disk cells apoptosis and ECM degradation by functioning as competing endogenous RNAs to bind to miR-200c and, thus, regulate the targeting gene XIAP (Cheng et al, 2018). There were also several other identified circRNAs reported to participate in the pathogenesis of IDD in the past few years (Li et al, 2019). However, it remains undetermined how circRNAs play a part functionally in the involvement of mechanical loads during the IVD degeneration development.…”

“…Accumulating evidence have revealed that non-coding RNAs, including microRNAs (miRNAs) and circular RNAs (circRNAs), functionally participate in the pathophysiological process of IDD (Li et al, 2015(Li et al, , 2019. MiRNAs are highly conserved, short, and single stranded endogenous RNAs which can regulate the target mRNA expression via associating with AGO proteins (Bartel, 2004).…”

“…ECM degradation, abnormal apoptosis of NP cells, and in ammatory reaction are the main pathological processes of IDD. ECM proteins are mainly composed of collagen II and aggrecan, which are mainly secreted by NP cells [12]. Therefore, the balance between the growth and apoptosis of NP cells is key to the balance of the ECM and the development of IDD.…”

“…At present, it is thought that the pathogenesis of IDD is mainly related to aging, mechanical stress, degradation of the ECM marked by progressive loss of aggrecan and collagen II, and genetic factors [2]. Among them, ECM synthesis and degradation caused by the abnormal proliferation and apoptosis of NP cells are considered to be key factors in the pathogenesis of IDD [3,4].…”
