Osteoarthritis (OA) is a prevalent degenerative joint disease whose pathogenesis remains unclear. The research aims to investigate the roles of Circ_0136474/miR‐127‐5p/MMP‐13 axis in OA. Differentially expressed circRNAs and miRNAs in OA cartilage tissue were screened out and visualized by R project based on RNA‐seq data and microarray data respectively. qRT‐PCR was carried out for detection of relative expression levels of Circ_0136474, miR‐127‐5p, MMP‐13 and other inflammatory factors and Western blot analysis was conducted to detect the protein expression level of MMP‐13. CCK‐8 assay and flow cytometry were conducted to determine cell proliferation and cell apoptotic ability respectively. RNA‐fluorescence in situ hybridization (RNA‐FISH) experiments were conducted to confirm the immune‐localization of the Circ_0136474 and MMP‐13 in human tissues. Targeted relationships were predicted by bioinformatic analysis and verified by dual‐luciferase reporter assay. Our findings revealed that the expression levels of both Circ_0136474 and MMP‐13 in OA cartilage tissue were significantly higher than that in normal cartilage tissue. Circ_0136474 could suppress cell proliferation by facilitating MMP‐13 expression and suppressing miR‐127‐5p expression in OA. Overexpression of miR‐127‐5p negatively regulated MMP‐13 expression to enhance cell proliferation. Our study demonstrated that Circ_0136474 and MMP‐13 suppressed cell proliferation, while enhanced cell apoptosis by competitive binding to miR‐127‐5p in OA, which may well provide us with a new therapeutic strategy for osteoarthritis.

The role of circular RNAs (circRNAs) in regulating cartilage homeostasis in osteoarthritis (OA) has been reported. However, the regulatory mechanisms of circRNAs in OA synovium remains basically unidentified. The current study intended to divulge the expression profile of circRNAs in OA synovium and investigate the possible molecular mechanisms of circRNAs in synovitis in OA through an integrated bioinformatics analysis. A total of 35 synovium samples were collected, including 17 from patients with knee OA and 18 from controls. circRNA sequencing was then carried out on five OA synovium samples as well as five controls to explore the expression pattern of the circRNAs. Real‐time quantitative reverse transcription‐polymerase chain reaction (qRT‐PCR) was done to confirm the manifestation levels of six differentially expressed circRNAs. Gene Ontology (GO) as well as Kyoto Encyclopedia of Genes and Genomes pathway (KEGG) enrichment analyses were done for differentially expressed circRNAs using the DAVID database to annotate the functions. The circRNA‐miRNA coexpression network was then created to estimate the probable molecular regulatory mechanisms of specifically expressed circRNAs in OA synovium. Total of 122 circRNAs were found to be differentially expressed in OA synovium through RNA sequencing. The expressions of five downregulated circRNAs as well as an upregulated circRNA were confirmed through the use of qRT‐PCR. The circRNA‐miRNA network was created to annotate the probable molecular regulatory mechanisms of specifically expressed circRNAs. Our outcomes revealed that circRNAs might be incorporated in the initiation as well as development of OA synovitis and might have prospective importance in OA diagnosis and therapy.

SummaryCircular RNAs (circRNAs) are a new class of RNAs that can be used as biomarkers in clinical blood samples. However, little is known about circRNAs' diagnostic values for rheumatoid arthritis (RA). In this study, the hsa_circ_0054189, hsa_circ_0008675, hsa_circ_0082689, hsa_circ_0082688, hsa_circ_0010932, hsa_circ_0002473 and hsa_circ_0044235 in peripheral blood were determined by quantitative reverse transcription–polymerase chain reaction (qRT–PCR). For hsa_circ_0044235, only one abnormal expression circRNAs in peripheral blood was selected as a targeted circRNA to explore the diagnostic value for RA. Our work demonstrated that the hsa_circ_0044235 in peripheral blood was decreased significantly in RA patients. The hsa_circ_0044235 in peripheral blood from RA patients did not correlate with C‐reactive protein (CRP), erythrocyte sedimentation rate (ESR), rheumatoid factor (RF), anti‐citrullinated protein antibodies (ACPA) or disease activity score 28 (DAS28). Receiver operating characteristic (ROC) curve analysis suggested that the hsa_circ_0044235 in peripheral blood has significant value in the diagnosis of RA. The risk score based on hsa_circ_0044235 in peripheral blood also distinguished significantly the patients with RA from systemic lupus erythematosus (SLE). This study suggests that the hsa_circ_0044235 in peripheral blood may be a potential biomarker of patients with RA.
