Circular RNA circZNF652 is overexpressed in osteoarthritis and positively regulates LPS-induced apoptosis of chondrocytes by upregulating PTEN

Abstract: BackgroundCircular RNA circZNF652 promotes LPS-induced in ammation, which contributes to the development of osteoarthritis (OA), indicating the potential involvement of CRNDE in OA. This study was carried to explore the involvement of circZNF652 in OA. Methods RT-qPCR was performed to analyze the expression of circZNF652 and PTEN mRNA in synovial uid samples from 60 OA patients and 60 healthy controls. Correlations between circZNF652 and PTEN mRNA were analyzed by Pearson's correlation coe cient. Overexpressio… Show more

“… 66 , 67 However, little is reported about its role in OA. Yuan et al 35 found that PTEN is overexpressed in OA and positively regulates the apoptosis of chondrocytes induced by lipopolysaccharide (LPS). Another study reported that PTEN negatively regulated Col2a1 and Aggrecan in chondrocytes.…”

“… 33 , 34 Recent studies showed that PTEN overexpression induced the apoptosis of chondrocytes and inhibited chondrocytes growth. 35 , 36 This study attempted to investigate the mechanism of USC-EVs in OA. We also explored whether hypoxic preconditioning could enhance the beneficial effects of USC-EVs on chondrocytes.…”

“…Many non-coding RNAs, including long-noncoding RNAs and circular RNAs, can regulate chondrocyte apoptosis through different mechanisms. Some of the mechanisms that can promote chondrocyte apoptosis involve Circ_0136474 [ 60 , 61 ], long non-coding RNA plasmacytoma variant translocation 1 [ 62 ], LINC00707 [ 63 ], and CircZNF652E [ 64 ]; the mechanisms that can prevent chondrocyte apoptosis include circANKRD36 [ 65 ], long noncoding RNA NAV2-AS5 [ 66 ], and circ_0020014 [ 67 ]. Circular RNAs act as sponges for miRNAs to suppress their activity and, thereby, regulate the expression of their downstream target genes [ 68 ].…”

“…The relationship between circRNAs and inflammation is now gaining recognition, and some studies have revealed that circRNAs participate in diabetes mellitus [ 241 ], atherosclerosis [ 242 ], osteoarthritis [ 243 ], and spinal cord injury [ 244 ], all of which are associated with immunoinflammatory effects. However, circRNA profiling and the regulatory mechanisms of neuroinflammation in PD remain unclear.…”
