Circular RNA Circ_0003221 Promotes Cervical Cancer Progression by Regulating miR-758-3p/CPEB4 Axis

Abstract: Background: Circular RNAs (circRNAs) play crucial roles in the development and progression of various cancers, including cervical cancer. However, the role and regulatory mechanism of circ_0003221 in cervical cancer are still unclear. Methods: The expression of circ_0003221, microRNA-758-3p (miR-758-3p), cytoplasmic polyadenylation element-binding protein 4 (CPEB4) was detected by quantitative real-time PCR (qRT-PCR). Cell Counting Kit-8 (CCK-8), colony formation, and 5-Ethynyl-2ʹdeoxyuridine (Edu) assays were… Show more

“…We attempted to grasp the potential molecular mechanism of circ_0005615 during CC progression. Online bioinformatics tool starbase 3.0 (http://starbase.sysu.EDU.cn/) was used to predict the possible miRNA targets of circ_0005615, and we selected eight miRNAs as the follow-up subjects due to their antitumor role in CC progression, including miR-149-5p, [13] miR-758-3p, [14] miR-205-5p, [15] miR-506, [16] miR-143-3p, [17] miR-138-5p, [18] miR-665, [19] and miR-331-3p. [20] RNA-pull down assay showed that miR-506, miR-138-5p, and miR-665 could be pulled down by circ_0005615 probe, especially miR-138-5p (Figure S2A).…”

“…Studies have shown that circRNAs can regulate the proliferation, metastasis, apoptosis and autophagy of various cancer cells by acting as ceRNAs 10,11 . For example, circ_0003221 accelerated cervical cancer progression by promoting CPEB4 expression through sponging miR‐758‐3p 12 . Meanwhile, circ_001422 could promote osteosarcoma progression through miR‐195‐5p/FGF2 axis 13 .…”

“…CircRNAs act as cancer regulators and novel biomarkers through sponging microRNAs (miRNAs) to affect gene expression [ 9 ]. Circ_0003221 accelerated cell malignant phenotypes of cervical cancer via regulating cytoplasmic polyadenylation element-binding protein 4 by playing the sponge effect on miR-758-3p [ 10 ]. Also, circ_0003221 was displayed to lead to the promotion of BLCA cell migration and proliferation [ 11 ].…”
