CircPRKCI relieves lipopolysaccharide‐induced HK2 cell injury by upregulating the expression of miR‐545 target gene ZEB2

Abstract: The aim of this study was to investigate the possible influences of circPRKCI abnormal expression on lipopolysaccharide (LPS)‐induced HK2 cell injury and its mechanism. The circPRKCI level was identified in serum samples from patients with urosepsis and healthy subjects, as well as LPS‐treated HK2 cells by qRT‐PCR. Cell viability, apoptosis, expression of proteins associated with apoptosis, and expression of pro‐inflammatory cytokines in LPS‐treated HK2 cells were measured. Effects of circPRKCI abnormal expres… Show more

“…In Hu's study of the mammary glands,he showed that circRNA-0001283 positively regulated HIPK3 expression by sponging miR-187. The results reveal a new functional circRNA-0001283 in breast cancer and may provide targets for developing novel therapeutic strategies for breast cancer [41]. “…A variety of endotoxins including LPS have been proved to stimulate the immune network response of the body to release inflammatory factors, and then lead to functional injury of kidney and other organs 24,25 . In addition, LPS‐induced HK2 cell injury has become a common in vitro cell model of septic AKI, providing convenience for performing the research on sepsis‐induced AKI 26 . Here, we investigated the circ‐ZNF644 effects on LPS‐treated HK2 cell injury to confirm its potential function in sepsis‐induced AKI.…”

Our data indicated that circ-ZNF644 had an increased expression in sepsisinduced AKI patients.…”

“…Meanwhile, it has been found that circPRKCI is a ceRNA to enhance AKT3 expression in esophageal squamous cell carcinoma via sponging miR-3680-3p [ 19 ]. CircPRKCI attenuates lipopolysaccharide-stimulated injury of HK2 cells by the up-regulation of ZEB2 by sponging miR-545 [ 20 ]. CircPRKCI depletion contributes to radiosensitivity and represses progression of esophageal cancer by the regulation of miR-186-5p/PARP9 axis [ 9 ].…”
