circCCT3 Enhances Invasion and Epithelial-Mesenchymal Transition (EMT) of Non-Small-Cell Lung Cancer (NSCLC) via the miR-107/Wnt/FGF7 Axis

Abstract: Background. CircRNAs play a role in a variety of biological processes, including tumorigenesis. circCCT3 has been shown to regulate cancer initiation and progression. Unfortunately, whether circCCT3 is involved in non-small-cell lung cancer (NSCLC) metastasis remains unclear. Methods. Our study utilized RT-qPCR to examine gene expression levels. A transwell assay was used to measure invasion ability of cells. Starbase software and TargetScan software were used to predict target genes. Results. circCCT3 knockdo… Show more

“…There have already been several studies that examine circRNAs as potential prognostic and diagnostic biomarkers in MM and investigate their mechanism of action 32,33 ; circCCT3 is such an indicative example. The oncogenic role of circRNAs from the CCT3 gene has also been demonstrated in other malignancies, such as non‐small‐cell lung cancer and pancreatic cancer 34,35 . Based on these, we aimed to investigate the expression levels of circCCT3 in plasma cell dyscrasias and shed light on its potential as a molecular biomarker in MM.…”

“…Wang et al found that the expression levels of GPC5 and WNT3a were negatively correlated in LUAD, and GPC5 could suppress the proliferation and metastasis of LUAD cells by inactivating the canonical WNT pathway by competitively binding to WNT3a [ 83 ]. In addition, miR-107 is another negative regulator of WNT3a, which inhibits the invasion and EMT of NSCLC cells by suppressing WNT3a and FGF7 expression [ 84 ]. In conclusion, these findings provide a theoretical basis for using WNT3a as a drug target to treat lung cancer.…”

“…In contrast, depletion of circ_0067934 hinders cell proliferation, migration, invasion and EMT and promoted apoptosis in NSCLC via inhibition of the Wnt/bcatenin pathway (79). Li et al demonstrated that circCCT3 functions as a sponge of miR-107 to enhance invasion and EMT of NSCLC via regulating Wnt pathway and FGF7 (80). In addition, circ_PRKDC (81) activate Wnt pathway to induce 5fluorouracil in colorectal cancer.…”
