Chrysin Ameliorates Cyclosporine-A-Induced Renal Fibrosis by Inhibiting TGF-β1-Induced Epithelial–Mesenchymal Transition

Abstract: Cyclosporine A (CsA) is a nephrotoxicant that causes fibrosis via induction of epithelial–mesenchymal transition (EMT). The flavonoid chrysin has been reported to have anti-fibrotic activity and inhibit signaling pathways that are activated during EMT. This study investigated the nephroprotective role of chrysin in the prevention of CsA-induced renal fibrosis and elucidated a mechanism of inhibition against CsA-induced EMT in proximal tubule cells. Treatment with chrysin prevented CsA-induced renal dysfunction… Show more

“…Studies have suggested that the occurrence of EMT could affect the cell migration in renal fibrosis procedure [ 23 ]. Western blot analysis was carried out to determine whether CCL17 affected EMT- or fibrotic-related proteins expression in HK2 cells.…”

“…In this study, β-catenin was not only the core target of all AO targets but also the most important target of AO in the treatment of AD. Among 13 phytochemicals of AO that bind to GSK3β, chrysin can also target Akt and MAPKs to affect protein phosphorylation ( Liu et al, 2016 ; Nagavally et al, 2021 ). Pinocembrin, which is a flavonoid, exerts neuroprotective effects through the GSK3β and Akt signaling pathways ( Kim et al, 2020 ).…”

“…Other anti-fibrosis signal pathways were also reported in the EMT process. For instance, EMT and glomerulosclerosis in rats with chronic kidney disease were suppressed by JAK/STAT signaling pathway ( Zhao et al, 2019 ), and cyclosporine-A-induced renal fibrosis was ameliorated by chrysin by the inhibition of TGF-beta-induced EMT ( Nagavally et al, 2021 ). In addition, the PI3K-Akt signaling pathway, a major regulator of anti-inflammation, has been reported to prevent renal fibrosis by attenuating renal tubular epithelial cell–mesenchymal transition ( Hu et al, 2021 ; Zhang Q et al, 2020 ; Wang et al, 2019 ).…”
