Chronic Sciatic Neuropathy in Rat Reduces Voluntary Wheel-Running Activity With Concurrent Chronic Mechanical Allodynia

Abstract: BACKGROUND Animal models of peripheral neuropathy produced by a number of manipulations are assessed for the presence of pathological pain states such as allodynia. While stimulus-induced behavioral assays are frequently used and important to examine allodynia (i.e. sensitivity to light mechanical touch; von Frey fiber test) other measures of behavior that reflect overall function are not only complementary to stimulus-induced responsive measures, but are also critical to gain a complete understanding of the e… Show more

“…In contrast to studies using exercise as a therapeutic tool, a number of studies have used voluntary running behavior as a diagnostic tool in persistent pain states [25; 43; 60; 61; 115; 127]. In studies using CFA-induced inflammation, wheel running in rodents pre-trained with running wheels is reduced for approximately 3 days post-injury [25; 60; 61].…”

“…However, this type of exercise-induced antinociception typically requires multiple weeks of daily wheel running [24]. Our previous studies [14, 15] and those of others [30] show that 5–8 days of baseline wheel running has no direct antinociceptive effect. These findings are consistent with human behavior in that consistent prolonged exercise can be used to maintain health, whereas pain or illness will disrupt a person’s exercise regimen.…”

“…The threshold for statistical significance was set a priori at α = 0.05 for all sets of multiple comparisons. In order to minimize unnecessary duplication, we used the minimum number of animals possible to make statistically significant conclusions, which was based on our previous publications [ 25 27 , 42 ]. These studies utilized sample sizes of N = 3 (IHC analysis) and N = 6 (behavioral analysis) to reliably produce statistically significant differences between treatment groups.…”
