Chronic lymphocytic leukemia B cells expressing AID display dissociation between class switch recombination and somatic hypermutation

Abstract: IntroductionAfter gene rearrangement, immunoglobulin (Ig) variable genes are diversified by somatic hypermutation (SHM), whereas the effector function of the constant domain is modified by class switch recombination (CSR). These processes depend on activation-induced cytidine deaminase (AID), a putative RNA-editing enzyme expressed in B cells from secondary lymphoid organs on CD40 ligand (CD40L) stimulation. 1 Given that the absence of AID expression in one form of the hyper-IgM syndrome in humans 2 and in AID… Show more

“…22 Our data support the suggestions regarding the association of AID expression and mutational status made in two pivotal observations. 28,30 However, the findings are in contrast to the recent report of Albesiano et al 31 These authors used more cycles for real-time amplification and found that all patients were PCR-positive. This underscores the necessity of standardization and the definition of appropriate threshold levels for AID.…”

“…[20][21][22][23][24] Activation-induced cytidine deaminase (AID) is normally expressed in CD19 þ /CD38 þ germinal centre B-cells and essential for somatic hypermutation, suggesting its potential association with IGV H mutational status. [25][26][27][28][29] Indeed, two reports have recently described the expression of AID in unmutated B-CLL cases. 28,30 However, a third study found no association between AID expression and IGV H mutation status.…”

“…13,25 Furthermore, the mutations of Ig genes in the preswitch region can be induced by modulating AID expression in CLL B-cells. 26 In contrast to these findings, we cannot exclude the possibility that a subset of genetically altered CLL cells acquire ability to express AID. This later theory is supported by the in vitro stimulation studies, where only a subpopulation of CLL cells is inducible to express AID.…”

“…Eight cases of CLL (cases [1][2][3][4][5][6][7][8] showed no transformation during the clinical course of the disease, 13 cases (cases 9-21) developed RT and eight cases (cases [22][23][24][25][26][27][28][29] showed PLT. In seven cases of RT (cases 9-15), both CLL and DLBL samples were available for analysis.…”
