Chronic Fluoxetine Reduces Serotonin Transporter mRNA and 5-HT1B mRNA in a Sequential Manner in the Rat Dorsal Raphe Nucleus

Abstract: In major depression in humans and in animal models of depression, there is a defect in serotonergic neurotransmission that can be relieved by chronic antidepressant treatment. One possibility is that this pathologic state is caused by excessive presynaptic autoreceptor activity in serotonergic neurons, and that antidepressants down-regulate the number of these inhibitory receptors, allowing more normal serotonin release to occur. To evaluate this hypothesis, we measured the effects of the antidepressant fluoxe… Show more

“…Both 5-HT 1A and 5-HT 1B receptors serve as autoreceptors in serotonergic cells and heteroreceptors in other neurons. 5-HT 1B autoreceptors are selectively regulated by fluoxetine [1,30] and chemical axotomy with 5,7-dihydroxytryptamine [31]. 5-HT 1A receptors are also regulated in a cell specific manner.…”

“…In this group of suicide victims, ethanol was detected in five suicide subjects and antidepressant drugs (clomipramine, fluoxetine, and mianserin) were detected in four depressed suicides. However, neither the above drugs nor electrocution or carbon monoxide influenced the transcription rate of adrenoceptors, serotonin receptors, or m-opioid receptors, except alcohol and fluoxetine that induced reductions in 5-HT 1B mRNA (Neumaier et al, 1996;Vazquez et al, 2002). In six suicides no toxicological screening data were available.…”

“…In brief, tissue sections were thawed at room temperature and fixed in cold 4% paraformaldehyde. After rinsing in phosphate-buffered saline, sections were treated with acetic anhydride (0.25% in 0.1 M triethanolamine), dehydrated, delipidated, and air-dried.In situ hybridization histochemistry for 5-HT 1A , 5-HT 1B , and SERT was performed using synthetic oligonucleotides as described previously (Neumaier et al, 1996, Neumaier et al, 1997, Neumaier et al, 2000, Clark et al, 2006. For the 5-HT 1B studies, three oligonucleotides corresponding to base pairs 1343-1382, 1630-1668, and 1790-1829 of the rat 5-HT 1B clone, MG11B (Hamblin et al, 1992) were used.…”
