Chronic Experimental Model of TNBS-Induced Colitis to Study Inflammatory Bowel Disease

Abstract: Background: Inflammatory bowel disease (IBD) is a world healthcare problem. In order to evaluate the effect of new pharmacological approaches for IBD, we aim to develop and validate chronic trinitrobenzene sulfonic acid (TNBS)-induced colitis in mice. Methods: Experimental colitis was induced by the rectal administration of multiple doses of TNBS in female CD-1 mice. The protocol was performed with six experimental groups, depending on the TNBS administration frequency, and two control groups (sham and ethanol… Show more

“…They induce naïve T lymphocytes to become Tregs (as shown in Figure 6 ). Our results match those obtained in the same model by Silva et al 2022 [ 61 ], and IL-10 therapy ameliorates TNBS colitis [ 62 ]. Moreover, Vegfa was also correlated positively to Il10 and Tgfb1 networks and negatively to SAA, the latter in the therapeutic approach.…”

“…Thereafter, 100 µL of 1% TNBS in 50% ethanol was administered through a catheter, carefully inserted until 4 cm into the colon. Then, mice were kept for 1 min in a Trendelenburg position to avoid reflux [ 18 , 29 ]. This procedure was performed on days 0, 7, 14, 21, and 28 of the experiment, over 5 weeks.…”

“…The most commonly used method to induce inflammation using TNBS in ethanol solution is a one‐time enema. In addition, repeated administration of TNBS can also induce chronic colitis, such as introducing 100 μL of 1% TNBS in 50% ethanol near the anus through a polypropylene catheter five times within 4 weeks, which leads to colon mucosal structure damage, intestinal wall thickening, and inflammatory cell infiltration similar to the symptoms of chronic CD (Silva, Solas, Pinto, & Mateus, 2022). It is noteworthy that chronic colitis induced by TNBS exhibits a persistent property of lamina propria fibrosis, which can be employed to investigate the pathological mechanism underlying the intestinal fibrosis accompanying human IBD (Wirtz et al., 2017).…”
