Chronic ethanol exposure inhibits distraction osteogenesis in a mouse model: Role of the TNF signaling axis

Abstract: Tumor necrosis factor-alpha (TNF-alpha) is an inflammatory cytokine that modulates osteoblastogenesis. In addition, the demonstrated inhibitory effects of chronic ethanol exposure on direct bone formation in rats are hypothetically mediated by TNF-alpha signaling. The effects in mice are unreported. Therefore, we hypothesized that in mice (1) administration of a soluble TNF receptor 1 derivative (sTNF-R1) would protect direct bone formation during chronic ethanol exposure, and (2) administration of recombinant… Show more

“…Chronic alcohol feeding is known to enhanced expression of pro-inflammatory cytokines including TNFα (Leudemann et al, 2005;Wahl et al, 2007). TNFα on the other hand exerts similar inhibitory effect on the expression of GLUT4 (Rotter et al, 2003).…”

“…A time course study of the serum TNF-a level (Fig. 3A) suggested that high levels of serum TNF-a would persist during the period (days 0-14) after tooth extraction, whereas the TNF-a level might be decreasing to a level <50 pg/ml during the late period after tooth extraction (days [14][15][16][17][18][19][20][21]. The significant reduction of serum osteocalcin level by LPS injection in WT mice appeared on day 7 not on day 21 after tooth extraction (Figs.…”

“…(10,17,18) However, there are few reports showing an inhibitory role for TNF-a in vivo or in clinical studies. A recent report showed that chronic ethanol exposure inhibited osteogenesis through the TNF signaling axis in a murine tibia distraction model, (19) but the mechanism by which TNF-a inhibits in vivo osteogenesis remains unclear. Clinical reports show that anti-TNF-a therapy increases BMD and plasma levels of osteocalcin in patients with rheumatoid arthritis, (20,21) but direct evidence proving the ability of TNF-a to inhibit osteogenesis is lacking.…”

“…While it has long been known that excessive alcohol consumption promotes adipogenesis and inhibits osteogenesis [5,6,7,8,9] and induces ER stress [11,12,13] that mediates alcoholic damages to many organs, the relationship between ER stress and osteogenesis disruption in BMSCs remained unknown. The present study provides evidence for the role of ER stress in mediating osteogenesis disruption after chronic alcohol exposure.…”

“…Excessive alcohol consumption substantially promotes adipogenesis and inhibits osteogenesis [5,6,7,8,9]. Moreover, both chronic and binge consumptions of ethanol are known to cause suppression of bone formation and enhancement of bone resorption.…”
