Chronic dexamethasone exposure retards growth without altering the digestive tract microbiota composition in goats

Abstract: BackgroundDexamethasone (Dex), an artificially synthetic cortisol substitute, is commonly used as an anti-inflammatory drug, and is also employed to mimic the stress state experimentally. It is well known that chronic stress disturbs the gut microbiota community and digestive functions. However, no relevant studies have been conducted in ruminants.ResultsIn this study, a low dosage of Dex (0.2 mg/kg body weight, Dex group, n = 5) was consecutively injected intramuscularly for 21 days to simulate chronic stress… Show more

“…There have been no publications focused on examining the relationship between cortisol and gut microbiota in humans. In goats, chronic dexamethasone exposure increased gut Prevotella [62], while in cattle, stress associated with transportation resulted in an initial increase and subsequent decrease of blood cortisol and a similar initial increase and subsequent decline in gut Prevotella , suggesting congruent changes in cortisol and Prevotella [63]. Previously, Prevotella was linked to psychiatric disorders prevalent in our population, including depression [64] and substance use disorder [65], while no studies evaluated association with post-traumatic stress disorder (PTSD).…”

“…Dexamethasone is a synthetic substitute for cortisol, commonly used as an anti-inflammatory and also used to simulate stress state. 61 Dexamethasone can alleviate joint swelling and pain by improving the expression of inflammatory cytokines and transcription factors, such as IL-1β, IL-17, TNF-α, and MMP-3 in primary synovial tissue cells of CIA rats. Studies have shown that dexamethasone can downregulate the expression of the NF-κB signaling pathway in the serum of CIA rats, effectively treat knee OA, stabilize oxidative stress, and relieve pain.…”

“…Finally, not all studies available show an effect of DEX in the GM. In a study performed with goats [ 107 ], chronic exposure to DEX did not change intestinal microbiota composition, suggesting that the GM was highly resistant to treatment with DEX.…”
