Chronic administration of isocarbophos induces vascular cognitive impairment in rats

Abstract: Vascular dementia, being the most severe form of vascular cognitive impairment (VCI), is caused by cerebrovascular disease. Whether organophosphorus causes VCI remains unknown. Isocarbophos (0.5 mg/kg per 2 days) was intragastrically administrated to rats for 16 weeks. The structure and function of cerebral arteries were assayed. The learning and memory were evaluated by serial tests of step‐down, step‐through and morris water maze. Long‐term administration of isocarbophos reduced the hippocampal acetylcholine… Show more

“…This condition results from the accumulation of excessive levels of acetylcholine in the synapses, glands, smooth, and skeletal muscles where cholinergic muscarinic and nicotinic receptors are found. 25 Excessive stimulation of these receptors in patients treated with rivastigmine, a dual BuChE and AChE inhibitor, comparing to patients treated with donepezil, a selective AChE inhibitor, can result in impaired cardiac and smooth muscles activity (manifested as bronchospasm, nauseas, or diarrhea), as well as fasciculations, myotonic jerks, weakness, and paresis. 25 Unfortunately, the latter symptoms can be misdiagnosed with neurological symptoms of advanced AD.…”

“…## p < 0.01 compared to the sham group; **p < 0.01 compared to the VD group. Reported studies reveal that the drug used for the management of Alzheimer's disease attenuates the cognitive function [1,12]. Data of this study suggest that zerumbone treatment ameliorates the learning and memory of the brain function and also significantly reduces the neurological function score compared to the VD group of rats.…”

“…Vascular dementia was induced in the rats as per previously reported method [12]. All the rats were anesthetized by administration of chloral hydrate (10% w/v) intraperitoneally (i.p.)…”
