Chromosome Sequence of Borrelia miyamotoi, an Uncultivable Tick-Borne Agent of Human Infection

Abstract: Borrelia miyamotoi is a newly recognized agent of human disease. B. miyamotoi strain LB-2001, an isolate from the tick Ixodes scapularis, was propagated in mice. The sequence of the chromosome was determined by next-generation sequencing of DNA isolated from whole blood. The sequence established that B. miyamotoi is a relapsing fever group species.

“…The procedures for the sequencing of genomes of Bmsl strain LB-2001, which was isolated from the blood of mice, and B. parkeri strain HR1 have been described (Barbour and Campeau Miller, 2014; Hue et al, 2013). DNA from a fresh culture harvest of B. anserina was extracted with Qiagen’s DNeasy Blood/Tissue Kit (Valencia, CA) and then treated with RNase I.…”

“…To provide additional material for phylogenetic inference, my laboratory carried out sequencing of LB-2001, a Bmsl strain and original isolate in North America (Hue et al, 2013), as well as the chromosome and much of the plasmid content of B. parkeri (Barbour and Campeau Miller, 2014). For the present study, these were added to existing chromosome and plasmid sequences of other Borrelia spp.…”

“…Until recently, for research purposes, B. miyamotoi was propagated by inoculation in SCID mice [87]. However, more recently, culture media based on modified Kelly-Pettenkofer medium were shown to The most relevant pathogens, ticks, geographic locations, clinical findings, and therapies are described.…”

“…To date, in order to identify protein families and dis-70 cover novel target antigen, salivary gland (SG) transcriptomes from 71 several ticks species have been described (Chmelar et al, 2008;72 Aljamali et al, 2009a;Francischetti et al, 2011). 73 Among the various transcriptome studies, next generation Williams-Newkirk et al, 2012;Hue et al, 2013;Kaewkong et al, 81 2014; Budachetri et al, 2014;Carpi et al, 2011) and parasitic 82 communities (Bonnet et al, 2014) associated with ticks.
