S U M M A R YDerivatives of Salmonella typhimurium carrying F prime or P prime plasmids with Klebsiella nif and his genes had specific nitrogenase activities similar to Klebsiella in selective conditions, even to showing ' hyperinduction ' under argon. No evidence was obtained for catabolite repression of normal nif expression but dibutyl cyclic AMP often augmented ' hyperinduction '. In non-selective conditions the Klebsiella his nifdeterminants were rapidly lost from the plasmids; the low levels of nifexpression and temperature-sensitive his expression previously reported were probably due to ready loss of his nifin the test conditions used.

In E. coli K12 (F'nif+ KP) hybrids, electron-transport-dependent phosphorylation is not necessary for anaerobic nitrogen fixation, and substrate level phosphorylation can provide sufficient ATP from glucose for nitrogenase activity, The fumarate-reduction system, however, is essential in these hybrids for the transfer of electrons to nitrogenase, This system is probably also involved in maintaining the membrane in the energized state, thereby allowing nitrogen fixation to occur, The nitrate-reduction system, which can energize the membrane like the fumarate-reduction system, is not necessary for nitrogenase activity in the E. coli KI2(F'nif+ Kp) hybrids, However, two nitrate reductase genes, chlA, and chlB, are essential for inhibition of nitrogen fixation by nitrate. Moreover, nitrate inhibits nitrogenase activity and this inhibition is most probably effected through a regulator factor coded by chlA and chlB,

In Klehsiellu pneumoniae, the physiological electron flow to nitrogenase involves specifically, in addition to nitrogenase reductase, the products of the nifF and nifJ genes. The J protein was purified to homogeneity and was found to be an iron-sulfur protein devoid of molybdenum. In its native state, the J protein is a dimer of M , about 245000, made up of two subunits of the same molecular weight. It contains about 30 mol iron and 24 mol labile sulfur/mol protein. The addition of J protein to crude extracts of a nifJ mutant reestablishes pyruvate-supported acetylene-reducing activity. This activity is further enhanced by addition of pure nitrogenase (Kpl). Based on its physical properties, the J protein is probably an oxidoreductase whose physiological role might be to transfer electrons from a metabolic donor to the F protein. In addition, another protein whose activity is also dependent on the nifJ gene seems to be required for the formation of a fully active Kpl.
