A simple, reliable and reproducible enzyme-linked immunosorbent assay (ELISA) using polyclonal antibodies for human cortisol binding globulin (CBG) has been developed. The sensitivity of the ELISA (1.20 fmol CBG/well) compared favourably with the sensitivity of other immunoassays. The excellent agreement (r = 0.98) seen between the present study and a binding assay indicates that the polyclonal antibodies used recognize only intact steroid-binding CBG. The intra- and inter-assay coefficient of variation (4.0% and 7.1% respectively) compared favourably with those reported by other authors.

A method for the determination of a-1-fetoprotein (AFP) in human serum by using a horseradish peroxidase (HRP) label in an enzymelinked immunosorbent assay (ELISA) is proposed. The method is based on the electrochemical determination of enzymatic reaction product with differential pulse voltammetry at a gold disk electrode. The assay consists of two successive steps. The ®rst step is a conventional HRP-mediated ELISA for the formation of electroactive 2,2 H -diaminoazobenzene by means of the o-phenylenediamine2 2H 2 O 2 2 2HRP system. At the second step, 2,2 H -diaminoazobenzene exhibits a sensitive voltammetric response at À0.19 V in pH 2.0 PBS. The peak current is proportional to the concentration of AFP in the range of 0.5±400 ngymL (R 0.9993) under optimum conditions. The sensitivity of this method is higher than that of the traditional spectrophotometric ELISA procedure. The proposed method has been applied to the clinical determination of AFP in human serum with satisfactory precision and accuracy.

Some basic components of an indirect competitive enzyme-linked immunosorbent assay (ELISA) were manipulated to reduce sulfathiazole (ST) detection limits. 3.3'.5.5'-Tetramethvlbenzidine fTMBj was more kffeitive than several aliemative chromogens tested fdr the hetection of horseradish peroxidase activity. Color development was maximized at pH 4.0. Using TMB and a 2M sulfuric acid stopping reagent, an assay was develoued using highlv diluted (l:SOO.OOO~ rabbit serum and low levels (o.oji3 pg/mY.,) 07 coating col;juga&, with a C value (inflection point of sigmoid standard curve) of 4.4 ppb ST. Urease was unsuitable as a marker for indirect competitive ELISAs.
