RESUMODesde a descoberta da primeira estatina, em 1971, os estudos dessa classe de fármacos evoluem sem parar, com resultados interessantes, que extrapolam sua ação farmacológica principal. As estatinas demonstram ter uma gama de ações farmacológicas intrigantes, os chamados efeitos pleiotrópicos, que incluem efeitos anti-infl amatórios, antidiabéticos, melhora da doença de Alzheimer, melhora da disfunção endotelial, antioxidante, entre outros. O objetivo desta revisão narrativa é demonstrar a quantidade de estudos realizados com esta classe de fármacos e confi rmar a relevância dos efeitos pleiotrópicos. Palavras-chave: Estatinas. Efeitos pleiotrópicos. ABSTRACTSince the discovery of the fi rst statin in 1971, studies of this class of drugs have been constantly evolving with interesting results that go beyond their primary pharmacological action. Statins are shown to have an intriguing range of pharmacological actions, the so-called pleiotropic effects, including anti-infl ammatory, antidiabetic, improvement in Alzheimer disease, improvement in endothelial dysfunction, antioxidant, among others. The purpose of this literature review is to demonstrate the amount of studies about this class of drugs and confi rm the relevance of pleiotropic effects. Keywords: Statins. Pleiotropic effects. IntroduçãoEm 1971, Akiro Endo iniciou o primeiro projeto com o objetivo de inibir a atividade da hidroxi--metil glutaril coenzima A redutase (HMGCoA redutase), enzima responsável pela síntese do colesterol. Em experimentos, a mevastatina tornou-se um potente inibidor, mas devido aos efeitos colaterais que exercia acabou não sendo comercializada (YAMMOTO; SUDO;ENDO;.Em 1990, depois dos avanços no estudo, entraram no mercado três fármacos derivados da mevastatina: lovastatina, sinvastatina e a pravastatina, atuando nesta via. Atualmente a classe das estatinas é amplamente utilizada na diminuição do colesterol (ENDO, 2010).As estatinas são agentes hipolipemiantes, inibidores seletivos e competitivos, responsáveis pela conversão da HMG-CoA redutase a mevalonato, um precursor do colesterol. A simple, sensitive and validated HPTLC method has been developed to determine Niacin and simvastatin simultaneously in synthetic mixture form. Chromatographic separation was achieved on a RP18 plate using a mixture of Methanol: Water: Acetic acid (60: 40:0.1) at a wavelength of 237 nm. Linearity of the method was found to be in the concentration range of 5000.0-25000.0 µg/ml for niacin and 100.0-500.0 µg/ml for simvastatin with correlation coefficient greater than 0.999. The method can be used for simultaneous determination of Niacin and Simvastatin.
