Chromatographic Study of Immobilized Boronate Stationary Phase

Abstract: The properties of a new polymeric material with immobilized boronic acid for HPLAC columns are discussed. In order to achieve a high boronic acid content, a support polymer was chemically bonded to polyamines, then phenylboronic acid groups were reacted onto the multiple reaction sites. The resulting support material showed a binding capacity of more than 1 meq/mL. A 4,6 x 54 mm analytical column was used to separate bioamines, nucleosides, and glycated hemoglobin effectively.

“…PBAs with strong and/or specific interaction with diols were first developed to improve purification and analytical techniques. They were notably used in purification columns with the aim to isolate biomolecules, including monosaccharides, nucleosides, and glycoproteins. These innovations inspired new technological developments for the sensing and release of small biological molecules, with application in drug delivery. , The more recent development of BE hydrogels has open a new era of research, with applications in local drug/cell delivery, 3D cell culture, and biofabrication.…”

“…This group of methods, known collectively as "boronate affinity chromatography", includes one of the earliest reported quantitative applications of affinity chromatography in the clinical laboratory-namely, the determination of glycohemoglobin for the assessment of long-term diabetes management ( Fig. 2) (7)(8)(9)(10)(11)(12)(13)(14)(15). At a pH above 8, most boronate derivatives form covalent bonds with compounds that contain cis-diol groups in their structure.…”

“…After the initial report by Mallia et al, similar low-performance methods were reported or evaluated by other groups (10 -13 ). The same approach has since been adapted for use in HPAC and HPLC-based systems (9,11,14,15 ).…”
