Chromatographic resolution and isotherm determination of (R,S)‐mandelic acid on Chiralcel‐OD column

Abstract: Resolution of racemic mandelic acid ((R,S)-MA) and numerical determination of binary competitive isotherm of (R,S)-MA on Chiralcel-OD column have been investigated in this study. The effects of the alcohol modifier and acidic additive in the mobile phase on the retention and enantioseparation of (R,S)-MA were studied at first. The inverse method was then used to determine the competitive isotherm parameters of (R,S)-MA by minimizing the sum of square deviations of the model predictions from the measured elutio… Show more

“…High-performance liquid chromatography (HPLC) is a versatile approach for both chiral analysis and manufacturing [2][3][4]. Enantioseparation can be achieved by the utilization of chiral mobile phases (CMPs) [5,6] or chiral stationary phases (CSPs) [7,8].…”

“…Enantioseparation has a particular emphasis in the pharmaceutical industry because a large number of examples of pairs of enantiomers exhibit differences in terms of bioactivity, pharmacological, and toxicological properties . The Food and Drug Administration and some other administrative authorities for drugs have recommended that chiral drugs must be tested and regulated separately .…”

“…Accurate equilibrium data is one of the most important keys to predict the overall SMB performance for chiral separation. Competitive isotherm parameters plus the rate coefficients have been determined from the single column experiments presented elsewhere . In addition, the apparent axial dispersion coefficient in each section of the SMB unit has been corrected with the real flow rate in that section based on the value obtained from the single column experiments .…”
