Chromatin structure and specificity revealed by immunological techniques

Abstract: As the techniques for preparing ‘clean’, well‐defined nuclei, soluble chromatin, well‐defined nucleosomes or polynucleosomes, and unfixed chromosomes improve the information obtainable by immunological techniques will be more exact and defined. Table 1 lists some uses of aerological techniques in the study of chromatin, chromosomes and their components.

“…The well-known relationship between troponin Ca 2+ affinity and cTnI phosphorylation at serine residues 22/23 led us to suspect elevated cTnI phosphorylation in Pkd 2+ / − hearts. 134 , 135 Western blots subsequently confirmed this hypothesis ( Fig. 4D ).…”

“…have been noted; e.g., the HI histones are both chemically and immunologically heterogeneous (Bustin & Stollar, 1972;Bustin, 1976). Antisera to HI histone subfractions showed little immunological correspondence between those of rat and calf thymus (Bustin & Stollar, 1973).…”

“…Most immunochemical studies on the structure and function of chromatin have used histone antibodies as analytical probes (Bustin, 1976(Bustin, , 1978(Bustin, , 1979. Such studies are based on the principle that the accessibility of histones at the surface of chromatin can be revealed with antibodies directed against antigenic determinants (or epitopes) of histone molecules.…”

“…© IRL Press Ltd., Oxford, England. 0261-4189/82/0104-0421$2.00.0 al., 1976;Absolom and Van Regenmortel, 1977). Since a 10-fold molar excess of peptide CN3 over intact H3 inhibited the binding in the H3 -anti H3 reaction by 8307, the antigenic activity of this fragment seemed particularly well preserved, and the corresponding epitope appeared a good candidate for further immunochemical investigations.…”
