The aims of this study were to estimate the ability of the Feulgen reaction to identify equine sperm nuclei with different ploidy levels, to determine the frequency of haploid, diploid and polyploid sperm nuclei in the semen of fertile equines and to evaluate the relationship between the nuclear DNA content and the nuclear area. Determination of the ploidy level of Feulgen-stained spermatozoa using a scanning microspectrophotometer was very similar to the subjective estimations made with a light microscope. This indicates that the Feulgen reaction is a simple, inexpensive and reliable technique to recognise the ploidy level of equine spermatozoa. The incidence of diploid and polyploid spermatozoa, determined with a light microscope in 11 fertile equines, was 0.17 ± 0.08% and 0.027 ± 0.027% respectively. DNA content values obtained by microspectrophotometry in the only equine that presented polyploid spermatozoa allowed us to discriminate between haploid, diploid and polyploid subpopulations. Measurement of the nuclear area discriminated only two subpopulations: one including the haploid and diploid subpopulations and the other including the polyploid one. The similarity between the area of the haploid and diploid sperm nuclei suggests that the increase in DNA content is anisotropic, with a privileged direction of growth perpendicular to the nuclear flattening plane.

RESUMOInvestigou-se a correlação entre a morfometria da cabeça e a intensidade da condensação e heterogeneidade da cromatina em espermatozoides de coelho (Oryctolagus cuniculus). Para tal, utilizaram-se 35 esfregaços de sêmen de coelhos da raça Nova Zelândia, corados com azul de toluidina e avaliados por análise de imagem computacional. As imagens foram obtidas digitalmente em tons de cinza e avaliadas por algoritmos desenvolvidos em ambiente de programação Scilab. As mensurações obtidas da cabeça dos espermatozoides foram área, perímetro, comprimento, largura, relação comprimento largura, elipsidade, fator de forma, descritores Fourier e simetria lateral e anteroposterior. Também foram avaliadas a intensidade da compactação e a heterogeneidade da cromatina espermática. Os espermatozoides de coelho apresentaram compactação e heterogeneidade cromatínica mais intensas do que os de touro e observou-se correlação significativa entre características morfométricas da cabeça e compactação e heterogeneidade cromatínica. An objective method for measuring bovine sperm nuclear shape was developed. Digital images of bovine sperm stained with propidium iodide were collected and Fourier functions used to describe the perimeters of individual sperm nuclei. Harmonic amplitudes from Fourier functions were first shown to be independent of sperm orientation during digitization. Sperm from 12 different bulls were used, and 6 harmonic amplitudes per sperm were found to adequately describe sperm nuclear shape. Based on harmonic amplitudes 0 to 5, cluster analysis was used to generate 20 different groups. Sperm within groups had similar morphologies and groups were distinguished by statistically unique shape characteristics. Harmonic amplitudes 0 to 5 can be used to distinguish previously reported abnormalities such as tapered, pyriform, macrocephalic, and microcephalic, as well as gradations in between. Furthermore, differences were detected among bull harmonic amplitude centroids (P < .05), indicating that bulls differ in mean sperm nuclear shape.
