“…The three drugs included in this study are known to undergo a variable degree of hydroxylation in the liver, via cytochrome-P450-dependent reactions; the main hydroxylated metabolites being p-hydroxyphenobarbital (Whyte & Dekaban 1977), 5-(4-hydroxyphenyl)-5-phenylhydantoin (Hoppel et al 1977;Perucca, Makki & Richens 1978) and 7-hydroxychlorpromazine (Kaul et al. 1972;Mackay, Healey & Baker 1974).…”

“…For flurazepam and its metabolites in blood and urine, the fluorometric procedure was based on hydrolysis to the benzophenones followed by K2CO3 cyclization to the 9acridanones (172). In the blood of schizophrenic patients, 11 chlorpromazine metabolites were quantitated as their DANSYL derivatives (439); the effects of pH, temperature, and solvent on the fluorescence of four phenothiazine derivatives were studied (671). A sensitive fluorometric method for protryptyline was found free of interference by other common psychiatric drugs (615).…”

“…A comparison of the methods used before 1972 has been given by Usdin (2) (for chlorpromazine and its metabolites only) and by Cimbura (3). Current methods with sufficient sensitivity include mass fragmentography (4), electron capture-GLC (5)(6)(7), and fluorescent derivatization (8)(9)(10). All these methods have some disadvantages; mass fragmentography is far too complex and the equipment too expensive to be considered for general purposes, electron capture-GLC can only be used when halogen atoms are present, and fluorescent derivatization only permits the detection of metabolites containing primary amine or hydroxyl groups.…”
