Chloroquine and hydroxychloroquine in the treatment of malaria and repurposing in treating COVID-19

Abstract: Chloroquine (CQ) and Hydroxychloroquine (HCQ) have been commonly used for the treatment and prevention of malaria, and the treatment of autoimmune diseases for several decades. As their new mechanisms of actions are identified in recent years, CQ and HCQ have wider therapeutic applications, one of which is to treat viral infectious diseases. Since the pandemic of the coronavirus disease 2019 (COVID-19), CQ and HCQ have been subjected to a number of in vitro and in vivo … Show more

“…Hydroxychloroquine has also been associated with reducing the risk of thrombosis, one of the major risk-factors in SARS-CoV-2 patients [103]. Although hydroxychloroquine is a major drug used both prophylactically and for the direct treatment of COVID-19 patients, but recently some adverse side-effects were reported, including cardiac arrest and ventricular arrhythmias [104][105][106]. Careful clinical examinations are still underway to validate the effects of hydroxychloroquine on COVID-19 patients.…”

“…The general information for some key drugs against COVID-19 are summarized in table 1. Most agents were originally approved for the treatment of other viral infections, except hydroxy-chloroquine that has been used for over 60 years primarily for malaria treatment, and is also approved for systemic lupus erythematosus [17].…”

“…Chloroquine (CQ) and its derivative, hydroxychloroquine (HCQ), are medications used in treatment and prophylaxis of the malaria and autoimmune diseases such as rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE) [ 9 ]. The only chemical structure difference between HCQ and CQ is that HCQ has a hydroxyl substitution at the ethyl group of amine ( Figure 1 ).…”
