Chloroplast DNA polymorphism in different types of cytoplasmic male sterile rice

Abstract: The lengths of open reading frame (ORF)100 and ORF29-TrnC GCA , the intronic sequence of rps16 and the transcribed spacer of TrnT UGU -TrnL UAA in chloroplast from different lines of cytoplasmic male sterility (CMS) rice were studied using indica types, japonica types and common wild rice as controls. The results show that the lengths of ORF100 and ORF29-TrnC GCA in CMS lines are similar to those of typical indica. The sequences of the rps16 intron and the TrnT UGU -TrnL UAA spacer in sporophyte sterile types … Show more

“…Additionally, it was found that the matK gene appeared three times in different primers of PCR amplification, and the amplification bands were enhanced in abortive buds, which showed upregulated expression, indicating that the abortion of radish buds had a great relationship with matK gene. Ou et al [ 77 ] studied the intron sequences of ORFs and rps16 genes in chloroplasts of different CMS lines in rice. Additionally, they found that in the gametophyte sterility line, the rps16 gene intron contains a GMTGAG sequence and a unique G at position 595, which can be used as a molecular marker to distinguish sporophytic sterility and gametophyte sterility.…”

“…Prentice et al (2003) implied chloroplast universal primers to study variation in cultivated and wild Prunus avium. Other examples for the use of universal primers to assess intraspecific variation are those carried out on Argania spinosa (Sapotaceae) of Morocco (El Mousadik and Petit 1996), Primula cuneifolia (Primulaceae) (Fujii et al 1999), Cannabis sativa (Kohjyouma et al 2000), the rare Spanish endemic, Silene hifacensis (Prentice et al 2003), Sagittaria latifolia (Dorken and Barrett 2004), and Oryza sativa (Ou et al 2009). …”
