Chitosan Hydrogel Doped with PEG-PLA Nanoparticles for the Local Delivery of miRNA-146a to Treat Allergic Rhinitis

Abstract: To prepare a binary formulation delivering miRNA-146 and evaluate a nucleic acid nasal delivery system by investigating its pharmacodynamic effects in allergic rhinitis. The gel/NPs/miR-146a thermosensitive in situ chitosan hydrogel carrying a nucleic acid was prepared and evaluated for its characteristics, including temperature sensitivity, gel strength, mucosal adhesion and drug release profile. After nasal administration of the formulation to ovalbumin-sensitized rats, the treatment of allergic rhinitis was… Show more

“…To date, several substances have been used to develop delivery systems for miR-146a and its mimics, including viral vectors such as adenovirus ( 162 ) and nano-carriers consisting of polyethylene glycol-poly (lactic acid) nanoparticles ( 163 ), cerium oxide nanoparticles ( 164 ) and polyethyleneimine nanoparticles ( 105 ). To promote absorption, the nanoparticles are frequently modified with lectins or penetrating peptides ( 165 ) or doped with chitosan ( 166 ). Although viral vectors possess good stability and transfection efficiency, they may induce adverse inflammatory responses.…”

“…20 Furthermore, RNAi-mediated gene therapy has excellent treatment specificity and can be conveniently carried out using various non-viral nanovectors, such as dendrimers, 14,21 dendrimer-incorporated hybrid nanoparticles, 22,23 and nanoscale formulations of solid lipid, 24 chitosan 25 and polylactic acid (PLA). 26 For instance, chitosan nanogels doped with polyethylene glycol-PLA can be used to deliver miRNA-146a for improved anti-inflammatory therapy of allergic rhinitis. 26 The miRNA-146a could effectively inhibit TNF receptor associated factor 6 (TRAF6) involved in the Toll-like receptor 4 (TLR4)/ myeloid differentiation factor 88 (MyD88) signaling pathway, thereby blocking the downstream nuclear transcription factorkappa B activation-induced pro-inflammatory cytokine overexpression.…”
