Chitosan-colloidal gold complexes as polycationic probes for the detection of anionic sites by transmission and scanning electron microscopy

Abstract: A new cationic colloidal gold complex has been developed for ultrastructural localization of cell surface anionic sites by transmission and scanning electron microscopy. The marker is prepared by labelling gold particles of suitable sizes (6 to 70 nm in diameter) with chitosan, a polymer of beta (1----4)-linked D-glucosamine. Using human red blood cells as a model, chitosan-gold complexes were shown to be specific for anionic sites and at pH 2 for sialic acid residues. The binding capacity of complexes of diff… Show more

“…In this case the cAu-polymer conjugate would act as a model drug delivery system. Chitosan-stabilised cAu has been successfully prepared by Horisberger and Clerc (1988) to use as a marker for nnionic sites on various micro-organisms and by Fiebrig et al (l994b, 1997) to visualize the sites of interaction of chitosan in a mucin-chitosan complex.…”

“…In candidae, N-carboxybutyl chitosan might also produce disturbances of membrane functions similar to those caused by polycations in other fungi (9). It is noteworthy that chitosan can agglutinate a number of bacteria, as well as mammalian cells (10), and chitosan-gold complexes have been used as polycationic probes for the detection of cell surface anionic sites (7). Moreover, polyelectrolyte and macromolecular complexes of chitosan with glycosaminoglycans have been reported (6,11), and alginic acid, which forms complexes with glycol chitosan (16), has been studied in conjunction with biotechnological applications of chitosan-alginate capsules (4,19).…”

“…Indeed, many macromolecules can be adsorbed on to colloidal gold while keeping their specific bioactivity. This has recently been reviewed (Horisberger, 1988). The phenomenon is com plex and depends on the stability of the colloid itself, the concentration, shape, configuration, and isoelectric point of the macromolecules, and the ionic strength, pH, and temperature of the suspending medium.…”
