Mucoadhesion is one of the most extensively studied methods for the administration of medication for faster onset of action with enhanced bioavailability. Present investigation was concerned with the development and characterization of controlled release muco-adhesive drug delivery system for the drug Tramadol Hydrochloride (TRD) to extend the duration of analgesia for patients suffering from acute to chronic pain. Formulations were made using the solvent casting technique, incorporating natural polymers like Guar Gum, Xanthum Gum and Chitosan in conjunction with HPMC (hydroxy propyl methyl cellulose) to modify the drug release. The compatibility of film components were validated by Fourier Transform Infra- Red (FTIR) spectroscopy and subjected to physico-chemical evaluation parameters and ex-vivo drug diffusion study. Optimized formulation was tested for analgesic efficacy in vivo to determine extent of their suitability in therapeutic use. All the prepared formulations exhibited superior pharmaceutical qualities with respect to drug content, folding endurance, swelling index and mucoadhesion time. In ex-vivo drug diffusion study, all the films displayed a controlled release lasting longer than eight hours. Formulations containing Chitosan in combination with HPMC (F6) showed better controlled drug release over other combinations. The formulation F6 bring out remarkable in vivo analgesic efficacy with 58.10 % analgesia in contrast to the standard (61.16%). It was deduced that Tramadol hydrochloride mucoadhesive films were found to be an effective treatment for moderate to severe pain with a quick onset and a lengthy half-life.

Over the last few decades, there have been significant strides in drug formulations and innovative administration methods, accompanied by a deepened comprehension of drug transport across tissues. Drug delivery encompasses a range of approaches, formulations, technologies, and systems designed to transport pharmaceutical compounds within the body, ensuring the safe and effective achievement of therapeutic effects. While often linked to a drug's chemical composition, it can also involve medical devices or combinations of drugs and devices. This concept is closely tied to dosage form and the chosen route of administration.These technologies and modes of administration play a crucial role in altering the drug release profile and influencing other pharmacokinetic and dynamic factors. The oral mucous membrane, known for its vascularity, high permeability, and accessibility, provides a means for the painless and steady systemic absorption of drugs. This approach bypasses the harsh stomach environment and initial liver metabolism. Buccal drug delivery, employing mucoadhesive polymers, has gained widespread application in this context.A variety of mucoadhesive devices, such as tablets, films, patches, disks, strips, ointments, and gels, have been recently developed. Among these, buccal patches stand out for their enhanced flexibility and comfort compared to other devices. This review sheds light on the novel drug delivery facilitated by mucoadhesive patches, delving into their historical evolution, manufacturing processes, properties, as well as examining the associated advantages and disadvantages.

Fluvoxamine is an antidepressant that functions pharmacologically as a selective serotonin reuptake inhibitor. Though it is in the same class as other SSRI drugs, it is most often used to treat obsessive-compulsive disorder. As the biological half-life of the drug is 15.6 hrs and belongs to BCS class II. To overcome these problems, Nanoparticles of Fluvoxamine were formulated by using Ethyl Cellulose, Eudragit RS 100 & Eudragit Rl 100 as a polymer by emulsification method. Among all the 9 formulations F6 formulation is optimized, as it shows maximum drug release at the end of 12hrs which suits the controlled release drug delivery system criteria as per our studies, having acceptable particle size, SEM, and Zeta potential value. From the drug release kinetics of the F8 formulation of Losartan Nanoparticles dispersion, it was concluded that the F8 formulation follows Zero-order drug release with super case II transport mechanism.
