“…Since the bioavailability of APIs is one of the crucial aspects in the formulation of orally administered dosage forms, it is the most frequently sought-after improvement in the development of nanostructures designed for this administration route. For this purpose, both polymeric/protein-based [72,[134][135][136][137][138][139] and lipidic nanostructures [131,140] have been employed and included in ODx.…”

“…Another study by Sinha et al used chitosan-alginate core-shell-corona shaped NPs to deliver DMF for long-term release, which had negative zeta potential of −27.2 mV and a bigger size of 561 nm, which lended it a larger surface area that was ideal for the matrix delivery system being investigated. This study, however, did not focus on the BBB and instead conducted various dissolution tests of the NP system to determine the best way to create an effective slow-release drug ( Sinha et al, 2021 ). These methods both show that existing therapies can also be improved using NP systems to deliver across the BBB more effectively.…”

“…An experimental design study was performed to optimize the composition of insulin-loaded nanoparticles prepared by ionic gelation of thiolated-N-dimethylethyl–CS conjugate with Na tripolyphosphate; the optimized nanoparticles were then embedded in a mucoadhesive CS-gelatin bilayered film [ 108 ]. CS-alginate core-shell-corona-shaped nanoparticles containing dimethylfumarate loaded in an orodispersible film were developed, which showed a 1.6-fold increase in AUC 0−4 and AUC 0−∞ values with respect to the conventional oral film formulation, even at a very lower drug dose (2 mg/film vs. 30 mg/film), allowing a reduction of drug dosage and of the related adverse effects in the treatment of multiple sclerosis [ 109 ].…”
