Chitin determination on marine seston in a shallow temperate estuary (Argentina)

Abstract: Chitin is one of the most abundant biopolymers in the planet. Chitin was quantified in seston fractions in order to obtain the first characterization of this polymer in the Bahía Blanca Estuary. Sampling was conducted at two sites: a non- impacted tidal channel -Bahía del Medio (BM)- and a sewage discharge sector -Canal Vieja (CV)-, during winter and summer. The highest values of chitin were observed in the seston fraction <20 µm (6.1 mg L-1 in BM in summer). The remaining larger fractions (20-60, 60-200, 2… Show more

“…The appropriate dye concentration may, however, depend on the chitin concentration in the water. We thus recommend 5 µg dye mL −1 , as chitin concentrations in natural waters are expected to be in the lower µg mL −1 range [ 69 ]. CFW-stained chytrids were difficult to distinguish from the background fluorescence on PC filters after staining with 1 µg CFW mL −1 , and even indistinguishable from the background at 5 and 25 µg CFW mL −1 .…”

“…The zooplankton chitin supply from the BBE was mainly presented in the form of degraded particles contributing to a micro-detritus pool in the seston size fraction ≤20 μm. (Biancalana et al, 2017a;2019). It is known that the carcasses and exuviae of zooplankton represent a concentrated reservoir of chitin for the water column bacteria and fungi (Tang et al, 2010).…”

“…m -3 ) during the warm season (Sabatini, 1989;Hoffmeyer, 2004;Dutto et al, 2012;Berasategui et al, 2016). Previous studies on chitin quantification on seston samples from the BBE suggested that most the chitin was micro-detritus from exuviae and pellets of A. tonsa during the warm season (Biancalana et al, 2017a;Biancalana et al, 2019). However, little is known about the production and stock of chitin by this copepod and its contribution to the seston in this ecosystem.…”

“…A set of samples from Bahia Blanca Estuary (BBE, Argentina) was used for developing the protocol. Water was pre-filtered with a 20 m net, in order to obtain the size fraction ≤20 m that previous studies had shown contains the major bulk fraction of chitin in certain marine environments, including BBE [11,22,30].…”
