CHIP functions as a novel suppressor of tumour angiogenesis with prognostic significance in human gastric cancer

Abstract: Decreased CHIP expression in GC resulted in increased angiogenesis and contributed to GC progression and poor prognosis. CHIP expression is a GC candidate clinical prognostic marker and a putative treatment target.

“…Similarly, CHIP has been found to colocalize with ataxin-1 nuclear inclusions [102, 103]. Coimmunoprecipitation experiments using cytoplasmic and nuclear fractions of mammalian cells showed that CHIP interacted with p65 [104], RUNX1 [105], and RUNX2 [106] proteins only in the nuclear extracts but not from the cytoplasmic fractions. Furthermore, under heat-stress CHIP not only assisted p53 to regain its native and transcriptionally active form but also comigrated with it into the nucleus and was found tethered to p21 and p53 promoters along with p53, as revealed by chromatin immunoprecipitation [107].…”

“…At the first, staining of CUL1 in the tissue was scored independently by two pathologists blinded to the clinical data, by applying a semiquantitative immunoreactivity score (IRS) which is reliable even in the hands of non-pathologists with a short training (Bai et al 2013;von Wasielewski et al 2002;Wang et al 2013). First, category A documented the intensity of CUL1 immunostaining as 0-3 (0, negative; 1, weak; 2, moderate; 3, strong) (Fig.…”

“…The standard protocol used for the immunostaining was provided as the previous study (Wang et al 2013). The monoclonal rabbit anti-CUL1 (1:200, Epitomics, California, USA), anti-cyclin A (1:100, Cell Signaling Technology, MA, USA), anti-cyclin E (1:200, Cell Signaling Technology, MA, USA), anti-p21 (1:200, Epitomics, California, USA), anti-MMP-2 (1:200, Cell Signaling Technology, MA, USA) and anti-MMP-9 (1:200, Epitomics, California, USA) were used for primary antibody incubation at 4 °C overnight.…”

“…Researchers have observed that IL-8 serves an important role in the prognosis of several types of carcinoma, including breast, colorectal, lung, gastric and prostate cancer (77,(104)(105)(106)(107)(108). Increased levels of IL-8 and MMP-3 are indicators of poor prognosis in triple-negative breast carcinomas (109).…”
