Chikungunya Virus Interacts with Heat Shock Cognate 70 Protein to Facilitate Its Entry into Mosquito Cell Line

Abstract: Aim: The study was designed to identify putative Chikungunya virus (CHIKV) receptor/s on C6/36 cells that facilitate viral entry. Methods: The virus overlay protein binding assay (VOPBA) was adopted to identify CHIKV-interacting bands present in C6/36 cell membrane and identity of the protein was established by mass spectrometry. The role of this protein as a putative CHIKV receptor on C6/36 cells was confirmed by infection inhibition assay. Cell surface localization of the identified protein was studied by in… Show more

“…The interaction between CypA and CD147 was further confirmed by Co-IP as described by Ghosh ( 18 ). Briefly, 100 μg protein lysates (RIPA buffer) were incubated with 20 μL of 50% Protein A/G-Agarose beads and a 2.5 μL mouse anti-CypA antibody (Abcam, ab58144, United Kingdom) or mouse anti-CD147 antibody (Abcam, ab666, United Kingdom) or 2.5 μL anti-IgG antibody at 4°C overnight.…”

“…Moreover, these flavonoids have also been reported as inhibitors of other single-stranded positive-sense RNA viruses. Apigenin, luteolin, kaempferol, and quercetin show inhibitory effects against the chikungunya virus (CHIK-V) [38,39,40]; apigenin, quercetin, and isorhamnetin show inhibitory effects against the hepatitis C virus (HCV) [41,42,43]; luteolin and quercetin show inhibitory effects against the dengue virus [39,44]; luteolin and kaempferol show inhibitory effects against the Japanese encephalitis virus (JEV) [45,46]; and isorhamnetin shows inhibitory effect against the Zika virus [47].…”

“…The list of proteins generated via LC‐tandem MS (MS/MS) analysis of the tryptic digested protein products obtained on matrix‐assisted laser desorption ionization time‐of‐flight mass spectrometry analysis were sorted using an algorithm (Ghosh et al , ). The proteins were arranged initially in the descending order of ‘scores’, number of unique peptides and peptide spectrum matches.…”
