e Both interleukin-17A (IL-17A) and IL-17F are proinflammatory cytokines that have an important role in intestinal homeostasis via receptor signaling. These cytokines have been characterized in chickens, but very little is known about their receptors and their functional activity. We provide here the first description of the sequence analysis, bioactivity, and comparative expression analysis of chicken IL-17RA (chIL-17RA) in chickens infected with Salmonella and Eimeria, two major infectious agents of gastrointestinal diseases of poultry of economic importance. A full-length chIL-17RA cDNA with a 2,568-bp coding region was identified from chicken thymus cDNA. chIL-17RA shares ca. 46% identity with mammalian homologues and 29.2 to 31.5% identity with its piscine counterparts. chIL-17RA transcript expression was relatively high in the thymus and in the chicken macrophage cell line HD11. The chIL-17RA-specific small interfering RNA inhibits interleukin-6 (IL-6), IL-8, and IL-1␤ mRNA expression in chicken embryo fibroblast cells (but not in DF-1 cells) stimulated with chIL-17A or chIL-17F. Interaction between chIL-17RA and chIL-17A was confirmed by coimmunoprecipitation. Downregulation of chIL-17RA occurred in concanavalin A-or lipopolysaccharide-activated splenic lymphocytes but not in poly(I·C)-activated splenic lymphocytes. In Salmonella-and Eimeria-infected chickens, the expression levels of the chIL-17RA transcript were downregulated in intestinal tissues from chickens infected with two Eimeria species, E. tenella or E. maxima, that preferentially infect the cecum and jejunum, respectively. However, chIL-17RA expression was generally unchanged in Salmonella infection. These results suggest that chIL-17RA has an important role in mucosal immunity to intestinal intracellular parasite infections such as Eimeria infection.

The immune system of birds diverged from that of mammals over 200 million years ago, and although the basic structure and function is largely similar, the immune system of birds is generally more compact and less polymorphic than that of mammals. Birds possess only a single polymorphonuclear leucocyte, the heterophil, but have cellular thrombocytes that may act as phagocytes and antigen‐presenting cells. Recognition of pathogens through pattern recognition is very similar to mammalian species. Within the adaptive immune response, birds have considerably less polymorphism of major receptor families. The most studied avian species, the chicken, has a compact major histocompatibility complex expressing only two Class I and Class II alleles and evidence of coevolution with major pathogens. There are only three immunoglobulin classes (IgY, IgM and IgA) but three T‐cell receptor classes. Functionally, the system is similar to mammals with cellular (Th1), humoral (Th2), regulatory and Th17 all described in the chicken. Vaccination is a widely used tool in disease control in chickens and other domestic birds. Vaccination technology is particularly advanced and includes the use of drinking water, spray and in ovo vaccines to facilitate delivery to large numbers of animals. Key Concepts Birds possess a complex immune system which, although shares much of its structure and function with mammals, has differences that reflect 200 million years of divergent evolution. Most of our knowledge of avian immunology comes from the domestic chicken and the development of vaccines for use by the poultry industry. B lymphocytes are named after the Bursa of Fabricius, the primary lymphoid organ where they mature in birds. Birds lack structured peripheral or mesenteric lymph nodes. The immune system of birds is more compact than that of mammals, with less polymorphism of major receptor families. Birds have three immunoglobulin or antibody classes compared to five in mammals. Birds possess three T‐cell receptor types, one more than mammals. Pattern recognition receptors such as Toll‐like receptors are largely conserved between mammalian and avian species. Birds possess a single polymorphonuclear cell class, the heterophil, but their nucleated thrombocytes appear to play a key role in innate immunity. The compact ‘minimal’ major histocompatibility complex of the chicken gives rise to close association between MHC type and resistance to infection. The need to vaccinate up to 50 billion animals reared each year has driven novel vaccines and delivery systems in the poultry industry.

Purpose The in vivo efficacy of ultrasonicated Rosmarinus officinalis ethanolic extract (UROEE) and its chitosan-loaded nanoparticles (UROEE-CsNPs) was investigated as a dietary prophylactic agent and as a therapeutic treatment against Eimeria tenella infected broiler chickens. Methods Chickens were infected with 4 × 104E. tenella oocysts at 21 days old for primary infection and with 8 × 104 oocysts at 35 days old for secondary infection. Eleven experimental groups were conducted. Dietary addition of 100 mg/kg UROEE and 20 mg/kg for CsNPs as well as UROEE-CsNPs were included for prophylactic groups from day 1 to 42. The same doses were used for therapeutic treatment groups for 5 constitutive days. Oocyst output in feces was counted. Histopathological and immunohistochemical studies were conducted. Gene expression of pro-inflammatory cytokines as IFN-γ, IL-1β and IL-6 as well as anti-inflammatory cytokines as IL-10 and TGF-β4 was analyzed using semi-quantitative reverse transcriptase-PCR. Results The results showed an efficacy of UROEE, CsNPs and UROEE-CsNPs in reduction of oocyst excretion and improving the cecal tissue architecture. CD4+ and CD8+ T lymphocytes protein expression were reduced. E. tenella infection lead to upregulation of pro-inflammatory cytokines as IFN-γ, IL-1β, IL-6 and anti-inflammatory cytokines as TGF-β4 following primary infection, while their expression was downregulated following secondary infection. Conclusion The dietary prophylactic additives and therapeutic treatments with UROEE, CsNPs and UROEE-CsNPs could decrease the inflammatory response to E. tenella as indicated by oocyst output reduction, histopathological improvements, CD4+ and CD8+ T cells protein expression reduction as well as reducing mRNA expression levels of the tested cytokines following primary and secondary infections. Consequently, these results will help to develop better-combating strategies for the control and prevention of coccidiosis on poultry farms as a dietary prophylactic agent or as a therapeutic treatment.
