Pharmaceutical research in natural products represents a major strategy for discovering and developing new drugs. The use of medicinal plants for the treatment of parasitic diseases is well known and documented since ancient times e.g. by the use of Cinchona succiruba (Rubiaceae) as an antimalarial. This chapter provides a comprehensive review of the latest results in the field of antiparasitic drug development from biologic sources (plants, bacteria, fungi and marine organisms) focussing on the treatment of protozoal infections (Plasmodium, Leishmania, Trypanosoma spp.). The status of validated in vitro and in vivo assays is reviewed, discussing their different features, problems and limitations. Because of the high number of natural products tested against the aforesaid protozoa in the last years, we limit the discussion to lignans, phenolics, terpenoids, and alkaloids as defined natural product classes. The review also covers essential research topics of recent publications on specific natural products (e.g. licochalcone A, benzyl-and naphthylisoquinoline alkaloids, and artemisinin) and gives an outlook to semisynthetic approaches of drugs already introduced in clinics or in clinical trial studies.

Mémoire.Un modèle original d'amibiase caecale mis au point chez le raton a été utilisé pour étudier l'effet de la levure Saccharomyces boulardii sur le développement des lésions. Des ratons infectés par Entamoeba histolytica ont été traités par Saccharomyces boulardii ; chez les traités, le nombre de ratons malades est significativement moins élevé, la gravité de l'affection, appréciée par l'aspect macros copique du caecum et la présence d'amibes a été diminuée. Les lésions observées sont semblables à celles des animaux témoins, mais leur processus de cicatrisation est accéléré. Par ailleurs, Sac charomyces boulardii n'a pas d'action amoebicide in vitro. Summary: Effects of Saccharomyces boulardii yeast on trophozoites of Entamoeba histolytica in vitroand on cecal amebiasis in the young rat.Lyophilized and rehydrated Saccharomyces boulardii yeasts were administered to young rats previously inoculated into the cecum with Entamoeba histolytica. The numbers of diseased young rats and the severity of the infection, assessed from the appearance of the cecum and its content of mucus and amebae, were signifi cantly reduced by the treatment. The lesions observed resembled those seen in the untreated controls, although healing was faster in the treated animals. Saccharomyces boulardii had no intrinsic amebicidal action in vitro.

RÉSUMÉ. Le Raton a été utilisé comme modèle expérimental d'amibiase caecale, en pratiquant l'implantation, sans effraction du caecum, d'une suspension de trophozoites d'Entamaeba histo lytica, souche gnotoxénique Rahman, de virulence élevée. L'atteinte de la paroi est objectivée par l'aspect macroscopique du caecum, par la densité des trophozoites présents et par un examen anatomopathologique. Comme il existe une corré lation entre les paramètres macroscopiques et l'histologie, un indice de gravité est défini, qui prend en compte plusieurs critères macro-et microscopiques.Ce modèle permet donc de suivre les différentes phases de la cicatrisation et de la guérison des ratons et d'apprécier ainsi l'activité de substances antiamibiennes. The young rat was used as an experimental model of caecal amibiasis by implan ting, without injury of the caecum, a suspension of E. histolytica trophozoïtes gnotoxenic strain Rahman of high virulence.The degree of pathogenicity is demonstrated by the macroscopic aspect of caecum, by the density of trophozoites and by an anatomopathologie examination. As a correlation exits between all these macroscopic parameters and histology, a degree of illness is fixed which include several macro-and microscopic criteria. This model enables to observe the different phasis of cicatriza tion and of recovering of young rats and so to appreciate the activity of antimoebic agents.
