The role of metabolites of arachidonic acid (AA) in experimental autoimmune uveoretinitis was studied using inhibitors of AA metabolism. Nordihydroguaiaretic acid (NDGA), which inhibits predominantly the lipoxygenase (LO) pathway, or indomethacin, a specific cyclo-oxygenase (CO) inhibitor, was administered to rats imunized with retinal S antigen. Levels of various AA metabolites were measured in the inflamed uvea, and the severity of intraocular inflammation was quantitated by morphometric analysis. Histopath-ologically, the uveoretinitis was significantly suppressed following treatment with NDGA, while indomethacin treatment resulted in augmentation of the disease (p < 0.05). These results tend to indicate that the inhibition of the LO rather than the CO pathway may be more beneficial in the treatment of autoimmune uveitis.

1 The inflammatory effects of hydroperoxy (HPETE) and hydroxy (HETE) acids, synthesized by arachidonic acid lipoxygenases, have been investigated in rabbit skin.2 High doses (10-20pg) of 5-, 12-or 15-HPETE or the 5,12-di-hydroxy acid, leukotriene B4 (0.1-1 fg), caused small but significant increases in plasma exudation following intra-dermal injection.3 Leukotriene B4 was equipotent with prostaglandin E2 and prostacyclin in potentiating bradykinin-induced plasma exudation, and was 100 times more active in this property than any other lipoxygenase product tested. 4 Leukotriene B4-induced plasma exudation was enhanced by prostaglandin E2. 5 The mono-HETEs were relatively inactive in causing or enhancing plasma exudation.6 Leukotriene B4 (0.1 fg) or prostaglandin El (1.0 pg) significantly elevated leukocyte accumulation in rabbit skin, whereas PGE2, 5-HPETE, 5-HETE, 12-HPETE or 12-HETE were inactive at doses up to 1 pg.

ABSTRACT-The effects of indomethacin on the production of cytokines at inflammatory sites were investigated in the monosodium urate (MSU) pleurisy model characterized by both cellular influx and edema. Indomethacin (10 mg/kg) orally administered 0.5 hr prior to MSU injection into the pleural cavity significantly inhibited MSU-induced neutrophil accumulation in the cavity. In addition, the drug slightly enhanced the level of MSU-induced tumor necrosis factor production without affecting interleukin-1 production. Furthermore, indomethacin inhibited both the levels of MSU-induced rat cytokine-induced neutrophil chemoattractant (CINC/gro) and interleukin-6 (IL-6) production by 78.3% at 3 hr and 45.8% at 4 hr post-injection, respectively. Although intrapleural injection of CINC/gro induced neutrophil infiltration in a dose-dependent manner, IL-6 did not affect the action of CINC/gro on neutrophil influx. These findings suggest that the inhibitory action of indomethacin on neutrophil infiltration is, at least, partly mediated by a decrease in the MSU-induced CINC/gro content in this model.
