Chemotactic factor-induced adherence of tumor cells

Abstract: Two peptides which have previously been shown to induce chemotactic motility in a number of tumor cells were tested for their ability to alter the adhesiveness of Walker Carcinosarcoma cells (A chemotactically-responsive rat tumor) and normal rat fibroblasts (which have previously been shown to be chemotactically nonresponsive). Adherence of the tumor cells to nylon fibers was increased in a dose-dependent manner by the two active peptides. Adherence of the fibroblasts was not increased. Nonchemotactic peptide… Show more

“…However, other types of studies show that the tumor cells respond very rapidly to stimulation with chemotactic factors. In both the cell swelling and foreign surface adherence assays, response to either the C5a-derived peptide or to FMLP can be detected within 1-3 min (16)(17)(18). This rapid response is similar to what is observed with leukocytes (12,13).…”

“…This procedure differs from the first in that the cells are in contact with the substrate for a much longer period of time (up to 24 hr). Both procedures have been described in detail in our recent reports (18,19).…”

These cells have been shown in previous studies to be responsive to phorbol esters in vitro and to chemotaetic peptides in vitro and in vivo [7][8][9][10]. Tissue culture reagents were purchased from Grand Island Biological Company (Grand Island.…”
